The increasing availability of large‐scale, complex data has made research into how human genomes determine physiology in health and disease, as well as its application to drug development and medicine, an attractive field for artificial intelligence (AI) approaches. Looking at recent developments, we explore how such approaches interconnect and may conflict with needs for and notions of causal knowledge in molecular genetics and genomic medicine. We provide reasons to suggest that—while capable of generating predictive knowledge at unprecedented pace and scale—if (...) and how these approaches will be integrated with prevailing causal concepts will not only determine the future of scientific understanding and self‐conceptions in these fields. But these questions will also be key to develop differentiated policies, such as for education and regulation, in order to harness societal benefits of AI for genomic research and medicine. (shrink)

Problem: Visions of technology, future scenarios, guiding visions represent imaginations of future states of affairs that play a functional role in processes of technological research, development and innovation—e.g. as a means to create attention, communication, coordination, or for the strategic exertion of influence. Since a couple of years there is a growing attention for such imaginations of futures in politics, the economy, research and the civil society. This trend concerns technology assessment as an observer of these processes and a consultant (...) on the implications of technology and innovation. TA faces increasing demands to assess imaginations of futures that circulate in the present and to participate in shaping these through scenarios or foresights. More than ever, this raises the question, which propositions can be made based on these imaginations by TA and how this can be used in advisory practices. Imaginations of futures are relevant for TA not as predictions but in their significance and effectiveness in the present, which need to be understood and assessed.Contents: This discussion paper outlines how present significance and effects of imagined futures in technological research and innovation processes can be conceived and analyzed. In this paper, all forms of imaginations of technology futures will be called “socio-technical futures” because within them technological developments and social changes are interwoven and inseparably interrelated. In this paper, we discuss why TA should analyze socio-technical futures, how such analyses can grasp the societal conditions that are expressed in the imagined futures and how these become effective in processes of technology development, communication, decision making etc. We raise the question which self-reflexive positioning or possible realignment of TA is needed as a response to its increased concern with assessing and even co-producing socio-technical futures. The latter is often demanded regarding the growing attention by politics and publics to imaginations of futures with wide temporal and spatial reach.Addressee of this paper is the TA community in a broader sense. The aim is to sensitize colleagues for the topic and its challenges, to consolidate discussions and to provide theoretical and methodical suggestions for research in TA and related advisory practices with respect to socio-technical futures. This paper has been originally initiated during the workshop “The present of technological futures-theoretical and methodical challenges for Technology Assessment”, in which all of the paper’s authors participated. Obesity and type 2 diabetes arise from a set of complex gene–environment interactions. Explanations for the heritability of these syndromes and the environmental contribution to disease susceptibility are addressed by the “thrifty genotype” and the “thrifty phenotype” hypotheses. Here, the merits of both models are discussed and elements of them are used to synthesize a “thrifty epigenotype” hypothesis. I propose that: (1) metabolic thrift, the capacity for efficient acquisition, storage and use of energy, is an ancient, complex trait, (2) the (...) environmentally responsive gene network encoding this trait is subject to genetic canalization and thereby has become robust against mutational perturbations, (3) DNA sequence polymorphisms play a minor role in the aetiology of obesity and type 2 diabetes—instead, disease susceptibility is predominantly determined by epigenetic variations, (4) corresponding epigenotypes have the potential to be inherited across generations, and (5) Leptin is a candidate gene for the acquisition of a thrifty epigenotype. Trent—as an erroneous path inspired by Augustine."—Robert Spaemann1Original Sin Passé—"after" Original Sin?The doctrine and later dogma of original sin had been a central topic of Catholic dogmatic theology up to the Second Vatican Council.2 One only has to consult the exhaustive bibliographies in the respective volumes Urstand, Fall und Erbsünde (in three parts) of the Handbuch der Dogmengeschichte.3 [End Page 693] Still echoing this long-standing, abiding preoccupation—researched during the Council and published in 1967—is the important tour d'horizon and noteworthy working hypothesis advanced by Henri Rondet, S.J., Original Sin: The Patristic and Theological Background.4 Soon after Vatican II a quite unexpected "sign of the time" found its way into Catholic theology—a widespread allergy to the theological topics of sin in general and of original sin in particular began to affect the post-conciliar mood, an unwillingness to acknowledge sin precisely as sin, let alone original sin and the concomitant original guilt. In polite and enlightened post-Vatican II Catholic theological society, one best ignored what was now seen as an unbecoming topic, unless the intention was, of course, to revise, that is, to "de-dogmatize," the received understanding of the Tridentine dogma of original sin, be it along Schleiermacherian lines (original sin as the sin of the world)5 or along Hegelian lines (the Fall and original sin as the unavoidable entailments of human finitude and freedom)6 or along Heideggerian lines (the co-conditionedness of the fundamental human existential situation of transcendental freedom by the historical facticity of collective human guilt since the inception of human history).7 Other post-conciliar progressive theologians took it upon themselves to go further than that and to "de-mythologize" the Catholic faith as a whole, and thus bring it up to date to the new standards of an [End Page 694] enlightened and increasingly secularized age: original sin, mortal sin, last judgment, hell, and eternal damnation all had to go in one or the other way together with angels, demons, and the evil one—at least the personhood (some sundry metaphysical principle of evil, possibly rooted in God, was, maybe, permissible; a person of a created, purely spiritual subsistence, definitely not)—into the dustbin of bygone theological notions, now thanks to a dynamic progressively transformative development of the understanding of faith, suddenly "untruths," or at best mythological remnants of a bygone pre-scientific age.8 Consequently, from the self-secularizing view of what took itself to be progressive Catholic theology, embracing the normative horizon of the immanent frame together with its immanent transcendence of perpetual human progress, the hamartiological and eschatological horizon brightened up considerably, especially in Germany, Holland, and Belgium, looking increasingly like the non-transcendent, historically immanent eschatology of Immanuel Kant's The End of all Things (1794)—a gradual vertical asymptotic approximation of human cultural and social progress to the ideal of the "kingdom of God." The problems that evolutionary theory and historical-critical exegesis of Genesis 1-3 might have posed for a de facto (salvation-)historical state of original justice became serendipitously immaterial, since the object of contention had been preemptively removed. There is a continuing irresolution on the levels, both of theory and political praxis, vis-à-vis a coming to terms with the problem of 'historical greatness'. This results from the pre-history of a concept which is, when seen in the context of a systematic theory of science, in two respects methodologically unsatisfactory. 1. The pre-idealist understanding of "greatness", in the sense of canonical exemplariness, is based on a timeless concept of morality, itself determined through a heteronomous concept of norm-giving transcendence and/or (...) nature. 2. The philosophy of history ignores the Kantian reference to the autonomy of the genesis of human orientation; instead it sees this genesis not as the expression of a potentially autonomous normativity but as a source of normative existential orientation. In this orientation the goal and the steps towards the goal are pre-reflexively implied. In the philosophy of history the 'historically great' function as genial mediators between telos and epoché. Their actions are not explicable by the categories of normal rationality. The lack of a sufficient methodology of socially practicable communication continues to lessen the chances of overcoming the practical and theoretical dependency on the idea of the mediating genius. Yet the shift to a critical social hermeneutic remains impossible as long as hermeneutic theory is willing to deny itself in the face of a purely scientistic analytic philosophy. (shrink)

Crusius est consideré comme l'un des plus importants critiques des théories philosophiques de Leibniz et de Wolff. Dans ce contexte, outre la critique de l'interprétation du principe de la raison telle que l'ont soutenue Leibniz et Wolff, il faut surtout citer le refus de l'utilisation de la méthode mathématique dans la philosophie. Le développement de la théorie des substances simples conçue par Crusius servira d'exemple pour démontrer le refus de cette méthode dans la métaphysique. La définition de la substance simple, (...) à laquelle Crusius parvient en ce fondant sur la distinction entre raison d'être et raison de connaître, s'offre à la pensée de Leibniz comme à la pensée de Wolff. Cependant, quand Crusius rejette l'interprétation de la substance simple comme point mathématique, il attaque surtout Leibniz. One of the two oldest manuscripts of Greek canon law, the Codex Patmiacus 172, sheds new light on the history of the Council in Trullo . Whereas from the other 45 extant manuscripts only 220 episcopal signatures to this Council have been known so far, the Patmiacus has preserved 6 additional signatures and thus raises the number of the hitherto known clerical participants to 226, representing a total of 227 bishoprics. Since these subscriptions are preceded by the signature of the (...) Emperor, the Patmiacus is the sole extant manuscript that bears in fact a total of 227 signatures, the number of participants assigned to this Council in the later canonistic tradition. Besides, this manuscript offers a great number of additions and corrections to the other 220 signatures. Due to the discovery of the six new episcopal signatures, however, the question of authenticity of the six position markers found in some manuscripts to indicate the place for the signatures of six episcopal sees which were present at the Sixth Ecumenical Council in 681/82 but apparently absent in 691/92 must be reconsidered. Whereas so far these markers have been considered original because it was only by their connumerating that the number of 227 could be achieved, this argument has become obsolete by the subscription list of the Patmiacus. Furthermore, the missing of the first three markers in the oldest extant manuscripts as well as their odd positions makes it very likely that they are not original but were interpolated at the times of the Seventh Ecumenical Council in order to assimilate the beginning of the subscription list of the Council of 691/92 to the one of 681/82 and thus substantiating Tarasios' claim that the Council in Trullo was a continuation of the Sixth Ecumenical Council. Whether the West and the metropolitan sees present in 681/82 - Rome, Thessaloniki, Sardinia, Ravenna and Corinth - were in fact invited as well to the Council in 691/92 is once again an open question and cannot be answered by reference to the manuscript tradition. Finally, the reasons for the western opposition to this Council are not so much to be sought in the form of the subscription list but more in the content of the canons and the Logos Prosphonetikos.
