The secretory system of plant cells sorts a large number of soluble proteins that either are secreted or accumulate in vacuoles. Secretion is a bulk‐flow process that requires no information beyond the presence of a signal peptide necessary to enter the endoplasmic reticulum. Many vacuolar proteins are glycoproteins and the glycans are often modified as the proteins pass through the Golgi complex. Vacuolar targeting information is not contained in glycans as it is in animal cells; rather, targeting information is in (...) polypeptide domains as it is in yeast cells. Several such domains have now been identified, but these show little or no amino acid sequence homology. We discuss the possibilities that targeting of protein to plant vacuoles may involve receptors as well as aggregation of protein at low pH. (shrink)

In Chinese body culture, the construct of qi 氣—literally translated as breath or energy—is at the heart of several programs of self‐cultivation, as well as other domains of bodily knowledge related to the subjective and inter‐subjective realm of everyday life. Also, among Chinese societies and communities, discourses on qi have assumed social significance in the milieus of politics, religion, and popular culture. Therefore, it appears to be the case that a concern for the qi experience is significant to both the (...) Chinese sensorium and its sociocultural context. However, while this category is ubiquitous, we are still left with the question of how people learn, elaborate, and make sense of it. By drawing from an in‐depth ethnographic study of a group of neidan qigong 內丹氣功 practitioners in Taipei, Taiwan, the aim of this article is twofold. To a greater extent, it investigates what role qigong, a mind‐body practice to cultivate and balance the vital energy, plays in the individual learning, elaboration, and understanding of the qi sensory experience. To a lesser extent, it considers this case study in the broader sociocultural context, in order to demonstrate that a first‐person approach to the qi experience can contribute to understanding the dynamics between embodied learning, consciousness, and society. In this article by way of reply, the author responds to the challenging comments on The Democratic Horizon provided by Michelman, Benhabib, White, Scheuerman and Laden. In response to Michelman, some reflections are propounded (1) on the function of judicial review, in order to alleviate the tension between two understandings of the mandate of the highest interpreter of the constitution as aimed at remedying either an occlusion of democratic authorship or a shortfall of agreement, and (2) on the need to (...) rethink how the authority of the supreme interpreter relates to the will of the people in a deeply changed historical context. In response to Benhabib, the author discusses the new limits of the accommodation of diversity in Fairburg, a fictional polity that expands the Rawlsian standard of ‘ reasonable disagreement’, and defends the normative relevance and democratic credentials of his notion of ‘ multivariate polity’. In response to White, after recalling the importance of putting the ‘democratic ethos’ at the center of a reflection on the democratic quality of complex societies where formal procedures often mask elitist substance, the author defends the ‘political, not metaphysical’ credentials of ‘openness’ (as well as of ‘exemplarity’ as the ‘upstream source’ of openness) and accepts a complementarity of ‘openness’ and ‘presumptive generosity’. In response to Scheuerman, the author restates his focus on the democratic ethos as a supplement to, not a replacement of, the reflection on democratic procedures and defends the conceptual diversity of governance from government. In response to Laden, the author highlights the diversity of contexts to which his own and Laden’s versions of ‘democratic justification’ are responding and defends a more moderate version of ‘openness’, which still keeps constitutional essentials and political values shielded from ongoing questioning. Finally, the author interprets the debate aroused by The Democratic Horizon as indicative of the persistent vitality of the Rawlsian legacy in the 21st century. (shrink)

Data access and data sharing are vital to advance medicine. A growing number of public private partnerships are set up to facilitate data access and sharing, as private and public actors possess highly complementary health data sets and treatment development resources. However, the priorities and incentives of public and private organizations are frequently in conflict. This has complicated partnerships and sparked public concerns around ethical issues such as trust, justice or privacy—in turn raising an important problem in business and data (...) ethics: how can ethical theory inform the practice of public and private partners to mitigate misaligned incentives, and ensure that they can deliver societally beneficial innovation? In this paper, we report on the development of the Swiss Personalized Health Network’s ethical guidelines for health data sharing in public private partnerships. We describe the process of identifying ethical issues and engaging core stakeholders to incorporate their practical reality on these issues. Our report highlights core ethical issues in health data public private partnerships and provides strategies for how to overcome these in the Swiss health data context. By agreeing on and formalizing ethical principles and practices at the beginning of a partnership, partners and society can benefit from a relationship built around a mutual commitment to ethical principles. We present this summary in the hope that it will contribute to the global data sharing dialogue. (shrink)

Digital technologies hold promise to modernize healthcare. Such opportunity should be leveraged also to address the needs of rapidly ageing populations. Against this backdrop, this paper examines the use of wearable devices for promoting healthy ageing. Previous work has assessed the prospects of digital technologies for health promotion and disease prevention in older adults. However, to our knowledge, ours is one of the first attempts to specifically address the use of wearables for healthy ageing, and to offer ethical insights for (...) assessing the prospects of leveraging wearable devices in this context. We provide an analysis of the considerable opportunities associated with the use of wearables for healthy ageing, with a focus on the five domains of intrinsic capacity: locomotion, sensory functions, psychological aspects, cognition, and vitality. We then highlight current limitations and ethical challenges of such approach to healthy ageing, including issues related to access, inclusion, privacy, surveillance, autonomy, and regulation. We conclude by discussing the implications of our analysis in light of current debates on the ethics of digital health, and suggest measures to address the identified challenges. (shrink)

Seeking a historical-philosophical approach to science teaching, narrative texts have been used as pedagogical tools to improve the learning experience of students. A review of the literature of different types of narrative texts and their different rates of effectiveness in science education is presented. This study was developed using the so-called Historical Narrative as a tool to introduce science content from a historical-philosophical approach, aiming to discuss science as a human construction. This project was carried out in a 9th grade (...) Physics class in K-12 school, in Rio de Janeiro, Brazil. The steps involved in constructing a Historical Narrative based on the controversy over animal electrical fluid between Luigi Galvani and Alessandro Volta is reported herein. Finally, qualitative research results of the activities inspired by this Historical Narrative are presented with the purpose of answering the research question: to what extent do Historical Narratives support and enhance discussions about the Nature of Science, through teaching the scientific content in a historical-philosophical approach with 9th grade students? The results indicate that Historical Narrative, based on historical episodes, is a good “door opener” to teach scientific content in a historical-philosophical approach, introducing discussions about the Nature of Science without neglecting the scientific content or simplifying the discussions about the NOS. The paper aims to present a reading of the question of Testimony rising in Derrida's later works (from Faith and Knowledge to Poetics and Politics of Witnessing): the experience of Testimony as the irreducible condition of the relation to the Other, of every possible link among living human singularities and, thus, of the thinking of a community to come. This thinking is able to divert the community from the economy grounding and structuring it within our political tradition governed by the (...) metaphysics of presence, which demands the sacrifice of the Other in its multiple theoretical and practical forms. We intend to read this proposal and to point out its rich perspectives by bringing it into the articulation of an ethical-political archi-writing. So we suggest going back to Derrida's early analyses of phenomenology and to De la grammatologie in order to present a reading of archi-writing as the irreducible condition of the relation to otherness and, thus, of the experience through which a living human singularity constitutes itself, a singularity different from the one our tradition compels us to think of within the pattern of the absolute presence to the self, free from the relation to the other. This article responds to the critique of productivist essentialism, which is the view that the human is the productive animal, made against Marx. The author argues against this view and holds that Marx introduces a dialectical account of human essence with the notion of species being in the 1844 Manuscripts, which he then develops in The German Idology. This account of essence includes a static and dynamic moment, and in capitalism, the dialectic of essence has resulted in the appearance of (...) the human as the productive animal. Finally, the author argues that Marx’s critique of production and dialectical account of human essence allow us to better think the possibilities for a post-work future. (shrink)

In “Toward an Ethic of Discussion,” Jacques Derrida returns to the controversy with Jonathan Searle to clarify his position but above all because he “would have wished to make legible the (philosophical, ethical, political) axiomatics hidden beneath the code of academic discussion.” I intend, in turn, to return to this text in order to find in it not only the conditions of an ethics of academic discussion but also of interpretation in a deconstructive perspective. In “Toward an Ethic of Discussion,” (...) in fact, it is possible to point out the necessity of a certain ethical treatment of the texts. In particular, how can we determine the conditions of an ethical use of quotation against the always possible manipulation of the text that quotation makes possible? I attempt to answer this question claiming that the reading protocol of deconstruction meets these conditions showing us at the same time the ethical conditions of scientific discourse in general, beyond any scientific claim of objectivity. Fitch's Paradox and the Paradox of the Knower both make use of the Factivity Principle. The latter also makes use of a second principle, namely the Knowledge-of-Factivity Principle. Both the principle of factivity and the knowledge thereof have been the subject of various discussions, often in conjunction with a third principle known as Closure. In this paper, we examine the well-known Surprise Examination paradox considering both the principles on which this paradox rests and some formal characterisations of the surprise notion, (...) crucial in this paradox. Standard formalizations of the Surprise Examination paradox in modal logic do not seem, at first glance, to depend on either factivity or knowledge-of-factivity, but we will argue that both factivity and knowledge-of-factivity play a key implicit role in the paradox. Namely, they are implicitly, perhaps unintentionally, used in order to simplify the definition of surprise. We analyze modal logical formalizations of three versions of the paradox concluding that the Surprise Examination paradox is the result of two flaws: the assumption of knowledge-of-factivity, and the over-simplification of the definition of "surprise" accordingly. By fixing these two flaws, the Surprise Examination paradox vanishes. (shrink)

The author proposes to read Documentalità constantly referring to Jacques Derrida’s work, which is one of its main sources. This comparative reading develops through a double move: on the one hand, it attempts to explain Derrida’s famous sentence “there is nothing outside the text” taking into account the criticism elaborated in Ferraris’ work. On the other hand, it suggests an understanding of the theory of document as a development of Derrida’s project of a “Grammatology as a positive science”. The articulation (...) of this twofold movement is found in Derrida’s theory of archiwriting, to which Ferraris refers: the liberation of the theory of writing from the linear phonetic-alphabetic ideal in view of a multidimensional writing, which can explain the functioning of our thinking and its articulation with the systems of inscription that the new technologies have made available. Emerging when scientific thinkers for the first time were connecting spirit to electricity and magnetism, the somnambulist became to certain Romantics a disclosure of the difficulty of harmonizing unseen and seen, agency and necessity. This problem prominently arose (...) in Anton Mesmer's late eighteenth-century experiments in animal magnetism, in which the sleepwalker proved a paradox: an opaque revelation of transparent currents and an unconscious vehicle of sophisticated thought. Romantics such as Coleridge embraced the somnambulist as an embodiment of these troubles involved in relating motion and meaning. Coleridge and those who responded to him saw in the hypnotized human this sobering possibility: the invisible requires the visible to make itself manifest while the visible needs the invisible to become significant. 1 This is sublime and sinister: to elevate a material form to spiritual power is transcendence; to sink the boundless into a body is grotesque.Here I explore the philosophical implications of animal magnetism, the place of the sleepwalker in the history of ideas, and the role of the hypnotized human in Romantic writers, especially Coleridge. In the early modern period, Paracelsus argued that a vital fluid courses through the cosmos. The human being draws this flow like a magnet attracts force. When the body pulls to itself evil effluvia, it falls ill. To return patients to health, Paracelsus placed magnets on painful areas. He hoped to extract the unwanted current and return it to the cosmic swirl. 2 In "magnetizing" spirit, Paracelsus opened a door for empirical inquiry into the invisible world. In 1600, William Gilbert claimed that magnets correspond to the earth: both are animated by universal attraction and repulsion. 3 By the end of the seventeenth-century, Newton proposed that things are impelled by a force that draws and repels. This paper intends to verify the extent and effectiveness of the transforming appropriation of the Derridean concept of ‘differance’ by Stiegler with respect to the problems that, according to Stiegler, make this creative critical operation necessary; in particular with respect to the most recent question concerning the possibility of thinking about and putting into practice a ‘neganthropological différance’ capable of facing the ecological crisis that today seems to threaten the very existence of life on earth. Technics and Time 1. to analyze the distinction between ‘vital difference’ and ‘noetic difference’ that constitutes the condition of possibility of the ‘neganthropological différance.’ In this perspective, the distinction proposed by Stiegler seems to re-propose the hierarchically oriented oppositional structure that characterizes metaphysical thought and in particular the opposition between man and animal, attributing to the human being the ability to free himself from the constraints of his biological-natural condition. Finally, the paper attempts to account for the repercussions of this approach on the very possibility of an effective response to the ecological crisis, concluding with a provocation regarding the role that theory can and must play with regard to such an urgent and far-reaching problem. This book shows how pragmatics and philosophy are interconnected, and explores the consequences and ramifications of this innovative idea, especially in addressing and solving the problem of breaking Grice's circle. The author applies philosophy in order to get to a better understanding of pragmatics, and pragmatics in order to get a better understanding of philosophy. The book starts with a chapter on the non-cancellability of explicatures and the role that this idea plays in the resolution of Grice’s circle, and proceeds (...) with the discussion of other topics in which explicatures or cancellability play an important and decisive role. While the reader proceeds in the reading of this book, they accumulate notions and pieces of knowledge which will be of invaluable use when arriving at the chapter on conversational presuppositions, where the author expresses his most radical views: namely that presuppositions are indeed cancellable, contrary to what many believe. In her book Specifications Grading, Linda B. Nilson advocates for a grading regimen she claims will save faculty time, increase student motivation, and improve the quality and rigor of student work. If she is right, there is a strong case for many faculty to adopt some version of the system she recommends. In this paper, we argue that she is mostly right and recommend that faculty move away from traditional grading. We begin by rehearsing the central features of specifications grading (...) and providing two examples of how to implement it in philosophy classes. In light of the examples, we argue that specifications grading fulfills two of Nilson’s central desiderata but not the third. Since specifications grading generates two benefits that when combined increase student learning, without adding or increasing burdens, we conclude that student learning increases when courses are revised to include aspects of specifications grading. Deliberative democracy has assumed a central role in the debate about deepening democratic practices in complex contemporary societies. By acknowledging the citizens as the main actors in the political process, political deliberation entails a strong ideal of participation that has not, however, been properly clarified. The main purpose of this article is to discuss, through Jürgen Habermas’ analysis of modernity, reason and democracy, whether and to what extent deliberative democracy and participatory democracy are compatible and how they can, either separately (...) or together, enhance democratic practices. Further exploration of this relationship will permit a better understanding of the possibilities and limits of institutionalizing both discourses, as well as of developing democracy in a more substantive dimension. Liberal democracy today, having aligned itself with capitalism, is producing a generalized feeling of weariness and disillusionment with government among the citizenry of many countries. Because of a decades-long march of globalized capitalism, economic oligarchies have gained oppressive levels of political power, and as a result, the economic needs of many people around the world have been neglected. It then becomes essential to remember that our ability to change society emerges from our power to formulate different questions; or, in this (...) case, alternative understandings of democracy. This book draws together a variety of alternative theories of democracies in a quest to expose readers to a selection of the most exciting and innovative new approaches to politics today. The consideration of these leading alternative conceptualizations of democracy is important, as it is now common to see xenophobic and racist rhetoric using the platform of liberal democracy to threaten ideas of plurality, diversity, equality, and economic justice. In looking at four different models of democracy this book argues that encounters with alternate conceptualizations of democracy is necessary if citizens and scholars are going to understand the constellation of possibilities that exist for inclusive, plural, economically equal, and just societies.
