Binding of a growth factor (GF) to its specific receptor on the cell surface causes the initiation of a signal transduction cascade which eventually results in mitosis. GF:receptor complexes are removed from the cell surface via receptor‐mediated endocytosis, a process which involves clathrin‐coated pits. After internalization into the endosomal compartment, a significant pool of GFs and GF receptors escape recycling to the cell surface and are sorted to the degradation pathway. The ligandinduced internalization and lysosomal degradation of GF receptors result (...) in the dramatic loss of surface receptors, a phenomenon termed receptor down‐regulation. In this review, we discuss relevant biochemical, morphological and kinetic studies of the mechanism of GF endocytosis, and the possible role of this process in mitogenic signaling by growth factor receptors. His respect for the author notwithstanding, Mendelssohn has little sympathy for Lessing's view of human (...) progress. Opposing the attempt to force the entire species into a single framework of linear development, Mendelssohn claims that, as a species, humankind has always oscillated between cultivation and barbarism. Borrowing the "ages of man" metaphor that Lessing employed in his own discussion, he writes that at almost any point in human history, man is "child, adult, and old man at the same time, though in different places and regions of the world." 1 Against Lessing's concept of global progress Mendelssohn proposes a model in which each individual follows his own trajectory of self-cultivation, a trajectory that begins before birth, intersects more or less briefly with the plane of historical time, and then continues after death into eternity.Mendelssohn's resistance to the Geschichtsphilosophie articulated here by Lessing and later appropriated by Kant, Schiller, and Hegel has been subject to a variety of interpretations. Not surprisingly, more recent commentators have stressed the philosopher's defense of particularity, his opposition to a totalizing theory of history that threatens to grant the individual nothing more than a bit part in the realization of some future state of human perfection. Norbert Hinske, for example, has argued that Mendelssohn's late writings engage in a conscious confrontation with Kant's understanding of historical progress. 2 [End Page 83]According to Hinske, especially Mendelssohn's short text On the Ideal Constitution indicates that the author "must have recognized spontaneously that Kant's proposed solution, whether intentionally or unintentionally, degraded the individual to a mere means of social progress...." 3Mendelssohn's concern for the individual is certainly central to his rejection of a species-based historical teleology, but one should also keep in mind the ethnic and cultural implications of such a model. As both Kant's and Hegel's later writings reveal, any attempt to understand human history in terms of a coherent, linear narrative, especially one that culminates in the rise of Christian Europe, is necessarily troubled by the continued existence of Judaism. In other words it is not just the individual who suffers an implicit reduction in status in late eighteenth-century theories of progress; entire cultures become vestigial once their historical function has been fulfilled. 4 Although the texts containing the most striking hostility toward Judaism were published after the appearance of Jerusalem, a somewhat surprising anti-Jewish bias is already evident in Lessing's explorations of the topic. Lessing does after all refer to the Jews in the period of Egyptian exile as "the most rude and the most ferocious" people imaginable. 5 Moreover, although he grants them a crucial role as educators of humanity, this role lasts only until the appearance of Jesus Christ, an event that simultaneously marks the end of the childhood of humankind. As Hans Liebeschütz has remarked, Lessing's adoption of this position must have been quite a blow to Mendelssohn. 6Each of these reasons for Mendelssohn's skepticism is significant in its own right; taken together, they also raise fundamental questions regarding his relationship to modernity and modern historical consciousness more generally. In one respect, of course, the topic of Mendelssohn's modernity has been much debated. Allan's monograph successfully takes on most of the (many) significant criticisms of the play, in particular those concerning its alleged disunity, and argues that the play is (...) more interesting than it has been taken to be. Allan sets out to show that the play merits more attention than it has received, and he succeeds, at least with this reviewer; a classicist reviewer who has written on the play is, however, hardly a test case. Moreover, the assertion of interest is not a strong claim, and, given the constraints of academic writing, scholarly arguments for a quirky play like the Andromache are often less compelling than the literary work itself. [End Page 126]Allan's thesis, that "every play represents a distinct thematic and dramatic complex which merits individual attention" (267), is undeniably true. He further asserts that the play is calculated to surprise and that the noticeable dissonances in it are purposeful as well. The book is divided into chapters on myth, structure, characterization, rhetoric, the spread of Attic tragedy, gender, the chorus, and the gods, in that order. Allan highlights Euripides' creative use of myth, drawing attention to the elevation of Andromache and the simultaneous way in which Euripides turns Neoptolemus, who was the center of epic treatments of the myth, into a corpse. There are connections between Allan's argument about the structure of the unexpected and Euripides' handling of myth. In part, Euripides builds the structure of surprise that Allan detects through his use of novel mythic variations.The elements of flexibility and surprise that Allan emphasizes also appear in a different guise in his considerations of gender (chap. 6) and the gods (chap. 8). According to Allan, female characters are useful for exposing the tensions that emerge when heroic society encounters crisis (162), while he uses Euripides' treatment of the gods to contribute to the overall argument about the multiplicity of Euripidean tragedy.Allan sounds a similar note in discussing characterization. Here he emphasizes dissonance, the contrast of traditional and invented story, and warns against expecting the depth of characterization typical of narrative from later centuries. This is a salutary reminder of the aesthetic conventions of tragedy too often overlooked by readers of the modern era.Characterization not only grows out of structure, as Allan argues, but is developed by rhetoric. His goal in the chapter on rhetoric is "to reappraise the functions and importance of Euripidean rhetoric (defined more broadly than the mere use of forensic topoi) by placing it within the larger dramatic context of the plays, and by viewing them within the wider rhetorical context of fifth-century Athenian culture" (119). Allan's contention is that Euripidean tragedy shows the failure of persuasion because of the intervention of political power. He argues that Euripides uses the Sophistic arsenal for his own dramatic ends, never for its own sake; as he notes, rhetoric is closely related to characterization through the characters' self-presentation. As he also shows, in the Andromache and other plays of Euripides, the rhetoric cannot be separated from the fictional world of the play. Ultimately, according to Allan, rhetoric reveals the feature of adaptability, sharing it with his other analytic categories of structure, myth and characterization.Allan asserts that there is a philosophical aspect to a plot based on surprise, but here I was disappointed and would have liked to hear more. We model a black hole spacetime as a causal set and count, with a certain definition, the number of causal links crossing the horizon in proximity to a spacelike or null hypersurface Σ. We find that this number is proportional to the horizon's area on Σ, thus supporting the interpretation of the links as the “horizon atoms” that account for its entropy. The cases studied include not only equilibrium black holes but ones far from equilibrium.

The transient cellular organelles known as migrasomes, which form during cell migration along retraction fibers, have emerged as a crutial factor in various fundamental cellular processes and pathologies. These membrane vesicles originate from local membrane swellings, encapsulate specific cytoplasmic content, and are eventually released to the extracellular environment or taken up by recipient cells. Migrasome biogenesis entails a sequential membrane remodeling process involving a complex interplay between various molecular factors such as tetraspanin proteins, and mechanical properties like membrane tension and (...) bending rigidity. In this review, we summarize recent studies exploring the mechanism of migrasome formation. We emphasize how physical forces, together with molecular factors, shape migrasome biogenesis, and detail the involvement of migrasomes in various cellular processes and pathologies. A comprehensive understanding of the exact mechanism underlying migrasome formation and the identification of key molecules involved hold promise for advancing their therapeutic and diagnostic applications. Ritual and Myth in the International Corona-Drama. A Conversation with Jeffrey Alexander.

Half of the 33.2 million people living with HIV today are women. Yet, responses to the epidemic are not adequately meeting the needs of women. This article critically evaluates how prevention of mother-to-child transmission (PMTCT) programs, the principal framework under which women's health is currently addressed in the global response to AIDS, have tended to focus on the prevention of HIV transmission from HIV-positive women to their infants. This paper concludes that more than ten years after their inception, PMTCT programs (...) still do not successfully ensure the adequate treatment, care and support of HIV-infected women. Of particular concern is the continued widespread use of single-dose nevirapine despite World Health Organization recommendations to employ more effective combination therapies that do not potentially jeopardize women's future treatment outcomes. In response, the article calls for a more comprehensive approach that places women's health needs at the centre of AIDS responses. This is critical in settings where the pandemic is generalized and there is a push to greatly expand PMTCT programs, as a more effective and equitable way of meeting the needs of women in the context of HIV. Without such a comprehensive approach, women will continue to be impacted disproportionately by the pandemic, and current strategies for prevention, including PMTCT, and treatment will not be as effective and responsive as they need to be. The production of a number of vaccines involves the use of cell-lines originally derived from fetuses directly aborted in the 1960s and 1970s. Such cell-lines, indeed sometimes the very same ones, are important to on-going research, including at Catholic institutions. The cells currently used are removed by a number of decades and by a significant number of cellular generations from the original cells. Moreover, the original cells extracted from the bodies of the aborted fetuses were transformed to produce the cell (...) lines, since otherwise they would be incapable of the kind of culturing that is required. "These notes are about the process of design: the process of inventing things which display new physical order, organization, form, in response to function." This book, opening with these words, presents an entirely new theory of the process of design. In the first part of the book, Christopher Alexander discusses the process by which a form is adapted to the context of human needs and demands that has called it into being. He shows that such an adaptive process will (...) be successful only if it proceeds piecemeal instead of all at once. It is for this reason that forms from traditional un-self-conscious cultures, molded not by designers but by the slow pattern of changes within tradition, are so beautifully organized and adapted. When the designer, in our own self-conscious culture, is called on to create a form that is adapted to its context he is unsuccessful, because the preconceived categories out of which he builds his picture of the problem do not correspond to the inherent components of the problem, and therefore lead only to the arbitrariness, willfulness, and lack of understanding which plague the design of modern buildings and modern cities. In the second part, Mr. Alexander presents a method by which the designer may bring his full creative imagination into play, and yet avoid the traps of irrelevant preconception. He shows that, whenever a problem is stated, it is possible to ignore existing concepts and to create new concepts, out of the structure of the problem itself, which do correspond correctly to what he calls the subsystems of the adaptive process. By treating each of these subsystems as a separate subproblem, the designer can translate the new concepts into form. The form, because of the process, will be well-adapted to its context, non-arbitrary, and correct. The mathematics underlying this method, based mainly on set theory, is fully developed in a long appendix. R. W. Sharples provides a new edition, with introduction and commentary in English, of the Greek text. The Mantissa is a collection of short discussions, transmitted as a supplement to the treatise On the Soul by the Aristotelian commentator Alexander of Aphrodisias (c.200 AD).The collection includes discussion of a range of topics, among them the nature of soul and intellect, theories of how seeing takes place, issues in ethics, and the nature of fate. The text is based upon a (...) new collation of the principal manuscript, the ninth century Venetus Marcianus graecus 258, and the apparatus corrects Bruns' misreportings of the principal manuscript and of the others that he used. Account has also been taken of the medieval Arabic and Latin versions of some of the sections which circulated independently, notably On Intellect which had a substantial influence on medieval philosophy. The introduction is chiefly concerned with the manuscripts and the relation between them. The commentary is based on the notes to the editor's English translation of the work (London: Duckworth and Ithaca: Cornell University Press, 2004); however, the commentary also takes into account more recent work on the collection by various scholars. This paper discusses two passages from Alexander of Aphrodisias’s commentary on Aristotle’s _ Topics _ that are transmitted in Ps-Jābir’s _ Kitāb al-Nukhab _. It argues that the Arabic translation of Alexander’s commentary may have been made from a fuller version than what came down to us in Greek. Especially since the author(s) of the Jābir-corpus form a tradition different from the school of Ḥunayn b. Isḥāq (d. 873) and authors associated to the ‘Baghdad school’, whose earliest figure (...) is Abū Bishr Mattā b. Yūnus (d. 940), the Arabic fragments of Alexander’s commentary preserved in the _ Kitāb al-Nukhab _ promise to shed more light on the early reception of the _ Topics _ and the different contexts in which Aristotelian dialectic was studied in the Islamic world. Originally the constitution was expected to express and channel popular sovereignty. It was the work of freedom, springing from and facilitating collective self-determination. After the Second World War this perspective changed: the modern constitution owes its authority not only to collective authorship, it also must commit itself credibly to human rights. Thus people recede into the background, and the national constitution becomes embedded into one or other system of 'peer review' among nations.This is what Alexander Somek argues is the (...) creation of the cosmopolitan constitution. Reconstructing what he considers to be the three stages in the development of constitutionalism, he argues that the cosmopolitan constitution is not a blueprint for the constitution beyond the nation state, let alone a constitution of the international community; rather, it stands for constitutional law reaching out beyond its national bounds. This cosmopolitan constitution has two faces: the first, political, face reflects the changed circumstances of constitutional authority. It conceives itself as constrained by international human rights protection, firmly committed to combating discrimination on the grounds of nationality, and to embracing strategies for managing its interaction with other sites of authority, such as the United Nations. The second, administrative, face of the cosmopolitan constitution reveals the demise of political authority, which has been traditionally vested in representative bodies. Political processes yield to various, and often informal, strategies of policy co-ordination so long as there are no reasons to fear that the elementary civil rights might be severely interfered with. A. G. Baumgarten's foundation of aesthetics as a branch of philosophical reflection is itself deeply rooted in Leibnizian metaphysics. From this starting-point Baumgarten comes to a kind of monadological concept of the work of art, that gets now an inexhaustible and thruthrepresenting meaning. This truth of the work of art however cannot be recognized rationally, for it is impossible for human beings to be aware of a beautiful object's 'metaphysical truth', which can only be perceived by the senses as - (...) to quote Baumgarten - 'aesthetical truth'. Still it is possible, by rational and scientific hermeneutics, to transfer some aspects of aesthetical to logical truth. The interest in art is therefore at the same time an interest in the cognition of the metaphysical cosmic order, and Baumgarten's aesthetics similarly are the first foundation of philosophical hermeneutics of art. The "Supplement" transmitted as the second book of "On the Soul" by Alexander of Aphrodisias is a collection of short texts on a wide range of topics from psychology, including the general hylomorphic account of soul and its faculties, and the theory of vision; questions in ethics ; and issues relating to responsibility, chance and fate. One of the texts in the collection, "On Intellect", had a major influence on medieval Arabic and Western thought, greater than that of (...) class='Hi'>Alexander's "On the Soul" itself. The treatises may all be by Alexander himself; certainly the majority of them are closely connected with his other works. Many of them, however, consist of collections of arguments on particular issues, collections which probably incorporate material from earlier in the history of the Peripatetic school. This translation is from a new edition of the Greek text based on a collation of all known manuscripts and comparison with medieval Arabic and Latin translations. (shrink)

Contemporary architecture is increasingly grounded in science and mathematics. Architectural discourse has shifted radically from the sometimes disorienting Derridean deconstruction, to engaging scientific terms such as fractals, chaos, complexity, nonlinearity, and evolving systems. That's where the architectural action is -- at least for cutting-edge architects and thinkers -- and every practicing architect and student needs to become conversant with these terms and know what they mean. Unfortunately, the vast majority of architecture faculty are unprepared to explain them to students, not (...) having had a scientific education themselves. Here is an architecture book by an architect/scientist, just in time to help architects in the new millennium. Alexander discusses many of the scientific terms arising in cutting-edge architecture, and explains them to those who don't have scientific training or advanced mathematical knowledge. We find discussions of the evolution of forms; the importance of process in design; iteration; genetic algorithms; sequences of transformations; different levels of scale (i.e. fractals); etc. They are explained here by an architect who is also a scientist, because he wants to change the way architects think and build. Alexander is not merely popularizing other scientists' results and making them accessible to architects: he is in fact presenting new and original scientific work that ties many of these concepts together in a way that will be useful to architects. There is an underlying assumption in the social sciences that consciousness and social life are ultimately classical physical/material phenomena. In this ground-breaking book, Alexander Wendt challenges this assumption by proposing that consciousness is, in fact, a macroscopic quantum mechanical phenomenon. The well-being of children is considered against the background of a wide variety of legal, political, medical, educational and familial perspectives. Some, notably Peter van Inwagen, in order to avoid problems with free will and omniscience, replace the condition that an omniscient being knows all true propositions with a version of the apparently weaker condition that an omniscient being knows all knowable true propositions. I shall show that the apparently weaker condition, when conjoined with uncontroversial claims and the logical closure of an omniscient being's knowledge, still yields the claim that an omniscient being knows all true propositions.

"Alexander's commentary on Book 1 concerns the definition of Aristotelian syllogistic argument; its resistance to the rival Stoic theory of inference; and the character of inductive inference and of rhetorical argument. Alexander distinguishes inseparable accidents, such as the whiteness of snow, from defining differentiae, such as its being frozen, and considers how these differences fit into the schemes of categories. He speaks of dialectic as a stochastic discipline in which success is to be judged not by victory but (...) by skill in argument.
