In contrast to the great majority of mycobacterial species that are harmless saprophytes, Mycobacterium tuberculosis and other closely related tubercle bacilli have evolved to be among the most important human and animal pathogens. The need to develop new strategies in the fight against tuberculosis (TB) and related diseases has fuelled research into the evolutionary success of the M. tuberculosis complex members. Amongst the various disciplines, genomics and functional genomics have been instrumental in improving our understanding of these organisms. In this (...) review we will present some of the recent key findings on molecular determinants of mycobacterial pathogenicity and attenuation, the evolution of M. tuberculosis, genome dynamics, antigen mining for improved diagnostic and subunit antigens, and finally the identification of novel drug targets. The genomics revolution has changed the landscape of TB research, and now underpins our renewed efforts to defeat this deadly pathogen. (shrink)

A woman is found lying dead on the floor of the living room of her house in Leiden, the Netherlands, and because of a swollen and a slightly wounded eyelid, an autopsy is performed on the body the day after it is found. Behind the wound, there is a whole ballpoint pen, which entered the head of the deceased through her right eye causing mortal brain damage. How did it get there? This question was to cause a stir in Dutch (...) society, holding a group of police detectives and several scientists in its grip for several years. In this article, the ballpoint case is analyzed as to the boundary work between credible and noncredible expertise. As it is often assumed that boundary work in continental law is preempted by the structure of these law models, this case study adds a comparative note to the growing literature about science and the law. The aim of psychology is to understand the human mind and behavior. In contemporary psychology, the method of choice to accomplish this incredibly complex endeavor is the experiment. This dominance has shaped the whole discipline from the self-concept as an empirical science and its very epistemological and theoretical foundations, via research practice and the scientific discourse to teaching. Experimental psychology is grounded in the scientific method and positivism, and these principles, which are characteristic for modern thinking, are still upheld. Despite (...) this apparently stalwart adherence to modern principles, experimental psychology exhibits a number of aspects which can best be described as facets of postmodern thinking although they are hardly acknowledged as such. Many psychologists take pride in being “real natural scientists” because they conduct experiments, but it is particularly difficult for psychologists to evade certain elements of postmodern thinking in view of the specific nature of their subject matter. Postmodernism as a philosophy emerged in the 20th century as a response to the perceived inadequacy of the modern approach and as a means to understand the complexities, ambiguities, and contradictions of the times. Therefore, postmodernism offers both valuable insights into the very nature of experimental psychology and fruitful ideas on improving experimental practice to better reflect the complexities and ambiguities of human mind and behavior. Analyzing experimental psychology along postmodern lines begins by discussing the implications of transferring the scientific method from fields with rather narrowly defined phenomena—the natural sciences—to a much broader and more heterogeneous class of complex phenomena, namely the human mind and behavior. This ostensibly modern experimental approach is, however, per se riddled with postmodern elements: creating phenomena in an experimental setting, including the hermeneutic processes of generating hypotheses and interpreting results, is no carbon copy of “reality” but rather an active construction which reflects irrevocably the pre-existing ideas of the investigator. These aspects, analyzed by using postmodern concepts like hyperreality and simulacra, did not seep in gradually but have been present since the very inception of experimental psychology, and they are necessarily inherent in its philosophy of science. We illustrate this theoretical analysis with the help of two examples, namely experiments on free will and visual working memory. The postmodern perspective reveals some pitfalls in the practice of experimental psychology. Furthermore, we suggest that accepting the inherently fuzzy nature of theoretical constructs in psychology and thinking more along postmodern lines would actually clarify many theoretical problems in experimental psychology. We would like to conduct a phenomenological analysis of illness beyond the characteristics which define it scientifically, in order to consider the suffering experience of the sick person which, in reality, constitute its authentic truth. Following Michel Henry’s philosophy which defines life as pathetic self-affection, we will understand that illness may and must first of all be considered as an intense form of this unsurmountable link to one’s self which concerns the very essence of everyone of us as a living (...) ipseity. Then, if “One does not feel”, if suffering is always a founding individual experience, the principle that medicine should be attentive to the subjective experience of patients is consequently established. Then we can also begin to understand the enigma of a patient’s attachment to his illness, to the point of refusing to be cured, as an inappropriate existential expression of an ontological fear of losing oneself. Researchers are increasingly expected to deliver “socially robust knowledge” that is not only scientifically reliable but also takes into account demands from societal actors. This article focuses on an empirical example where these additional criteria are explicitly organized into research settings. We investigate how the multiple “accountabilities” are managed in such “responsive research settings.” This article provides an empirical account of such an organizational format: the Dutch Academic Collaborative Centres for Public Health. We present a cross-case analysis of four collaborative (...) research projects conducted within this context. We build on Miller’s notion of “hybrid management.” The article shows that the extended concept of hybrid management is useful to study the different accountabilities encountered in such settings. We analyze how the collaboration developed and which conflicts or dilemmas arose. We then focus on the different hybrid management strategies used in the collaboration. The empirical material shows how the different aspects of hybrid management feature in various configurations in the four projects. We highlight that hybrid management strategies may be used by different groups or at different moments, may reinforce or contradict each other, and may be more or less effective at different points in time. The contribution focuses on Neo-Confucian texts as collected by Zhu Xi and Lü Zuqian and is a look from the ‘outside’, from the perspective of German theories of Bildung. It aims at demonstrating that among other insights that today’s readers may gather from Neo-Confucian literature, one aspect protrudes from others: that learning can be considered as a virtue—even a meta-virtue—a form of life and mode of self-formation of the person. It does not seem exaggerated, from this perspective, to state that (...) Neo-Confucian philosophy is—to a large extent—a philosophy of learning and self-transformation which offers fruitful irritants for questioning the widespread habits of thinking about skills and their development in today’s strong and problematic discourses and corresponding educational policies. Brain functions are stochastic processes without intentionality whereas mind emerges from brain functions as a Hegelian “change from quantity”, that is, on the order of 1012 profusely interconnected neurons, “into a new quality”: the collective phenomenon of the brain's self-experience. This self-referential and self-observing quality we have in mind is capable of (recursively) observing its self-observations, i.e., interpreting change that is meaningful in relation to itself. The notion of self-interpretation embodies the idea of a “hermeneutic circle”, that is, (in interpretation (...) theory) the treatment of the whole in relation to its parts. (shrink)

Two major management philosophies of the 20th Century, scientific management and quality management, are often contrasted. Scientific management is seen as a system that focuses on task efficiencies whereas quality management is described as a collaborative, people-centered process approach to continuous improvement. This paper examines the ethical implications of these diverse approaches, particularly in the way information is used to decide which employees to lay off in times of economic difficulty. The paper uses case examples of quality management as teaching (...) tools to place the conduct of managers within utilitarian, deontological and justice approaches to ethical decision making. Finally, it suggests that a third system, open-book management, may help deal with this ethical dilemma, though not without risk. (shrink)

Many writers have been struck by what Ronald Melzack, a leading investigator of pain mechanisms, calls the ‘puzzle’ of pain. Thus the surgeon Leriche, often quoted in this connection, says: Defence reaction? Fortunate warning? But as a matter of fact the majority of illnesses, even the most serious, attack us without warning. Sickness is nearly always a drama in two acts, of which the first takes place, cunningly enough, in the dim silence of bur tissues, with the lights out, before (...) the candles have been lit. When pain makes its appearance, we are almost in the second act…… Pain merely makes more distressing and sad a situation already long lost…… In fact, pain is always a baleful gift, which reduces the subject of it, and makes him more ill than he would be without it. Previous research described different cognitive processes on how individuals process distributional information. Based on these processes, the current research uncovered a novel phenomenon in distribution perception: the Endpoint Leverage Effect. Subjective endpoints influence distribution estimations not only locally around the endpoint but also influence estimations across the whole value range of the distribution. The influence is largest close to the respective endpoint and decreases in size toward the opposite end of the value range. Three experiments investigate this phenomenon: Experiment 1 (...) provides correlational evidence for the Endpoint Leverage Effect after presenting participants with a numerical distribution. Experiment 2 demonstrates the Endpoint Leverage Effect by manipulating the subjective endpoints of a numerical distribution directly. Experiment 3 generalizes the phenomenon by investigating a general population sample and estimations regarding a real‐world income distribution. In addition, quantitative model analysis examines the cognitive processes underlying the effect. Overall, the novel Endpoint Leverage Effect is found in all three experiments, inspiring further research in a wide area of contexts. (shrink)

We assess St. Paul's account of kenōsis in Philippians 2:5–8 from a neurophenomenological horizon. We argue that kenōsis is not primarily a unique event but belongs to a class of experiences that could be called kenotic and are, at least in principle, to some degree accessible to all human beings. These experiences can be well analyzed, making use of both a phenomenological approach and the cognitive neuroscience of altered states of consciousness. We argue that kenotic experiences are ecstatic, in that (...) they involve—both phenomenologically and neurologically—one's "stepping out of" his/her self and history. This seemingly impossible task of stepping out has led to the understanding of kenōsis as a unique event. We conclude that kenotic experiences are continuous with common, everyday experiences of the self's intimate communion with everything that exists. This means that kenotic Christology does not necessarily have to rest solely on the scriptures but can also be arrived at by way of the worldly experiences of actual, living persons. (shrink)

Is it permissible to kill an innocent person against her will in order to prevent several other innocent persons from being killed against their will? The answer to which this essay comes after extensive discussion is – under certain conditions and limitations – affirmative. On the way to this answer, the book offers a comprehensive in-depth discussion of so-called deontic restrictions – that is, the idea of an action’s being prohibited in circumstances in which performing it once would be the (...) only way to prevent its being performed multiple times. The book’s leading question is whether there is a plausible rationale for deontic restrictions. To this effect, a taxonomy and critical discussions of the most important approaches to justify deontic restrictions are provided – where many of these approaches undergo a deeper examination for the first time ever. In addition, the book clarifies some adjoining questions, such as why deontic restrictions are often perceived as being problematic or how the concept of agent-relativity should best be understood and formalized. Put into broader perspective, the conclusions offered should have a bearing on a number of debates in normative ethics, not least on the debate between deontologists and consequentialist. (shrink)

One of the constants in Noam Chomsky’s philosophical, linguistic and ethical positions is the existence of what he calls “human nature”. Following Marx, Darwin and last century’s revolutions in the social sciences, human nature has been one of the most contested conceptual holdovers from modern European philosophy. Chomsky’s discoveries and models on syntax and language make up one of the frameworks to most critically offset the traditional moral dimension of human nature. Contrary to most traditions prior to his work, language (...) can no longer be restricted to either mind, soul or spirit. Language, as Chomsky has continually upheld and sharply refined, is a physical and biological process. But how his notion of human nature derives from this process is complex, as he seems to disregard philosophy’s classic analytic delineation between the descriptive causal realm of human nature and the normative axiological extensions of the same concept. In this paper, we seek to examine the philosophical and ontological implications of Chomsky’s claim that human nature derives from the innate dimension of the language faculty. Not only does Chomsky maintain the category of human nature, he also indexes it to the question of freedom. We thereby argue for the coherence of his proposal and show how it operates to weld the perspective of a modal theory of biologically-rooted creativity to innate conditions specific to his theory of language generation. However, we question whether its restriction to humans alone is sustainable from a scientific perspective by putting forth the claim that Chomsky’s science is in fact a radical ontology of social subjectivation. Le langage, comme moyen d’expression de la croyance religieuse, entretient un malentendu : il apparaît comme un discours sur des vérités d’une supériorité telle que le croyant peut s’autoriser à faire des jugements d’ordre éthique ou esthétique envers – ou à l’encontre – d’autres personnes. Pourtant, loin d’être un discours logique sur des faits de connaissance, le discours sur la croyance religieuse est un jeu de langage qui manifeste une adhésion à une forme de vie.

Working with Old High German and Old Saxon texts has become rare in German language and literature studies. Nowadays, the brief introductions to historical linguistics and philology do not suffice to enable students to explore the oldest German texts in significant depth. A new tool could at least help move closer to this dept of textual exploration: an on-line publication of all Old High German and Old Saxon texts with a morphological word-by-word annotation. The data has been collected by the (...) research project ‘Old German Reference Corpus’ (‘Referenzkorpus Altdeutsch’ / ReA) and can already be used for complex database queries. Within the framework of the ‘eHumanities Centre for Historical Lexicography’ (eHumanities-Zentrum für Historische Lexikographie / ZHistLex), it has been converted into a clearly arranged format, focusing on the information needed for comprehending the texts: the ‘Lesekorpus Altdeutsch’ (LeA). This tool can of course not replace the work with text editions, translations and commentaries, but will help pave the way from the first insight into the language to a profound understanding. Additionally, a supplemental both-way linking between every word token and the corresponding entry in the on-line dictionaries of historical German is being prepared. (shrink)

In Nozick’s rendition of the decision situation given in Newcomb’s Paradox dominance and the principle of maximum expected utility recommend different strategies. While evidential decision theory seems to be split over which principle to apply and how to interpret the principles in the first place, causal decision theory seems to go for the solution recommended by dominance. As a reply to the CDT proposal by Wolfgang Spohn, who opts for “one-boxing” by employing reflexive decision graphs, I will draw on the (...) framework of causal knowledge patterns, i.e., Bayes net causal models, augmented by non-causal knowledge, to finally arrive at “one-boxing” – more intuitively and more closely to what actually is in Nozick’s story. This proposal allows the careful re-examination of all relevant concepts in the original story and might cast new light on the following questions: How may causality in general be understood to allow causal inference from hybrid patterns encoding subjective knowledge? How can the notion of prediction be analyzed – philosophically and formally? And what’s the decision-maker’s conceptualization of the situation he will act upon? (shrink)
