While large portions of the mammalian genome are known to replicate sequentially in a distinct, tissue‐specific order, recent studies suggest that the inactive X chromosome is duplicated rapidly via random, synchronous DNA synthesis at numerous adjacent regions. The rapid duplication of the inactive X chromosome was observed in high‐resolution studies visualizing DNA replication patterns in the nucleus, and by allele‐specific DNA sequencing studies measuring the extent of DNA synthesis. These studies conclude that inactive X chromosomes complete replication earlier than previously (...) thought and suggest that the strict order of DNA replication detected in the majority of genomic regions is not preserved in non‐transcribed, “silent” chromatin. These observations alter current concepts about the regulation of DNA replication in non‐transcribed portions of the genome in general and in the inactive X‐chromosome in particular. (shrink)

Deciphering how DCAFs (DDB1‐CUL4 Associated Factors) modulate a broad spectrum of cellular processes, including cell cycle progression and maintenance of genomic integrity is critical to better understand cellular homeostasis and diseases. Cells contain more than 100 DCAFs that associate with the Cullin‐Ring Ubiquitin Ligase 4 (CRL4) complex that target specific protein substrates for degradation. DCAFs are thought to act as substrate receptors that dictate the specificity of the ubiquitination machinery (“catalytic DCAFs”). However, recent studies have suggested that some DCAFs might (...) play a different role by targeting CRL4 complexes to distinct cellular compartments (“structural DCAFs”). Once localized to their correct cellular domains, these CRLs dissociate from the structural DCAFs prior to their association with other, substrate‐specific catalytic DCAFs. Thus, we propose that DCAF switches can provide a mechanistic basis for the degradation of proteins that regulate cell growth and proliferation at precise points in space and time. (shrink)

_Purpose_ of the research is to study the originality of interpretation of death in the lyrics of Ukrainian diaspora in Canada in the context of the opposition "foreign land – motherland", based on its existential development in philosophical anthropology and culture of the last two centuries. Its implementation presupposes, first of all, analysis of the forms of development and disclosure of the death motif by figurative and artistic means. _Theoretical basis__._ The author uses the well-founded tradition of interpreting the death (...) motif in philosophical literature of the last centuries, i.e. non-classical and modern philosophical thought. _Originality._ The present study is an attempt at systematization of the notions of death in Ukrainian poetry in Canada in the second half of the 20th century. It is based on the study of artistic texts and the worldview of Ukrainian emigrants. Appeal to the fiction of the Ukrainian Diaspora allows illustrating the originality of philosophical understanding of death in the context of the national culture. _Conclusions_. Reference to Ukrainian-Canadian poetry allows expanding the space of philosophical understanding of death in the form of figurative fiction. Concretizing the character of their interpretation of the motif of death, it is necessary to focus attention on its two basic forms of manifestation: existential, associated with living abroad, and sacrificial death for the benefit of the homeland. Thus, the motif of death occupies a significant place in the lyrics of representatives of the Ukrainian diaspora. The concept of human death is closely connected with the concept of life, which is concretized through their meaningful comprehension. This is a matter of interpretation of despair and loneliness motifs, as well as the illumination of the images of a foreign land, which is one of the features of emigrant literature in general. Many different writers have employed what might be termed the ‘existence criterion’ when offering realist analyses of perception. In all these realist accounts, a basic argument may be detected. If any experience—e.g. the having of either a sensory or a mental image—is to be a form of perceiving, then the statement reporting that experience must satisfy a condition which applies to all perceiving-statements. Any putative perceptual statement of the form ‘S perceives x’ must entail a statement to the effect that (...) x, or some y, does exist as a public perceptual object in S’s perceptual range. For example, my statement ‘I see Peter’ will be a proper perceptual statement only if Peter is a public perceptual object now in my line of vision, or only if I take some other public perceptual object now in my line of vision to be Peter. Statements about our having images of things, this basic argument continues, cannot satisfy this condition. The object imaged —e.g. Peter—is not always present in the subject’s perceptual range at the time of the imaging, and the subject does not necessarily take a public perceptual object in his perceptual range to be that object. Hence, any statement reporting our imaging something must be false if it is construed as a report of a perceiving. (shrink)

Open peer commentary on the article “Homeostats for the 21st Century? Simulating Ashby Simulating the Brain” by Stefano Franchi. Upshot: It is a mistake to characterise Ashby’s view of life (from the 1950s) as passive, abstractly modelled in part by the homeostat; one should distinguish the stasis of homeostasis from the activity of the (model) organism. Likewise mistaken is the accusation of contingency; one should distinguish the purposeless mechanism from the purposeful (model) organism. There is no basic conflict between Ashby’s (...) view and later developments in a similar tradition; technical advances are not the same as foundational gaps. (shrink)
