Deamination of adenosine in RNA to form inosine has wide ranging consequences on RNA function including amino acid substitution to give proteins not encoded in the genome. What determines which adenosines in an mRNA are subject to this modification reaction? The answer lies in an understanding of the mechanism and substrate recognition properties of adenosine deaminases that act on RNA (ADARs). Our recent publication of X‐ray crystal structures of the human ADAR2 deaminase domain bound to RNA editing substrates shed considerable (...) light on how the catalytic domains of these enzymes bind RNA and promote adenosine deamination. Here we review in detail the deaminase domain‐RNA contact surfaces and present models of how full length ADARs, bearing double stranded RNA‐binding domains (dsRBDs) and deaminase domains, could process naturally occurring substrate RNAs. The translational research community, in general, and the Clinical and Translational Science Awards (CTSA) community, in particular, share the vision of repurposing EHRs for research that will improve the quality of clinical practice. Many members of these communities are also aware that electronic health records (EHRs) suffer limitations of data becoming poorly structured, biased, and unusable out of original context. This creates obstacles to the continuity of care, utility, quality improvement, and translational research. Analogous limitations to sharing objective data in (...) other areas of the natural sciences have been successfully overcome by developing and using common ontologies. This White Paper presents the authors’ rationale for the use of ontologies with computable semantics for the improvement of clinical data quality and EHR usability formulated for researchers with a stake in clinical and translational science and who are advocates for the use of information technology in medicine but at the same time are concerned by current major shortfalls. This White Paper outlines pitfalls, opportunities, and solutions and recommends increased investment in research and development of ontologies with computable semantics for a new generation of EHRs. Thomas Spence was a native of Newcastle upon Tyne who is best known for his political writings, and more particularly for his radical `Plan' for social reform involving common ownership of the land. One hitherto neglected aspect of Spence's Plan was his proposal to extend the benefits of reading and of `correct' pronunciation to the lower classes by means of a phonetic script of his own devising, first set out and used in Spence's Grand Repository of the English Language. (...) The Grand Repository was one of many English pronouncing dictionaries produced in the late eighteenth century to satisfy the growing demands for a clear guide to `correct' pronunciation. It differs from its contemporaries firstly in that it was intended primarily for the lower classes, and secondly in that it is the only eighteenth-century pronouncing dictionary of English to use a truly `phonetic' script in the sense of one sound = one symbol. In this fascinating and unique account, Beal pays particular attention to the actual pronunciations advocated by Spence and his contemporaries with a view to reconstructing what was felt to be `correct' pronunciation in eighteenth-century Britain. With broad appeal to linguists and historians alike, this study highlights the importance of pronouncing dictionaries as a resource for the historical phonologist, and provides a valuable addition to the limited body of knowledge on eighteenth-century pronunciation. The classic theory of Common Law is based on the idea of a law derived from immemorial customs that guide judges’ decisions and contribute to the continuity and stability of the legal order. Hobbes, in his criticism of Edward Coke, questions the legal principles that characterize the “spirit of Common Law”. In his view, it is authority and not use that makes the law. This Hobbesian criticism, which can be considered as one of the ­sources of positivist interpretation of customary (...) law, first raises the question of the legal validity of custom but also leads to an original analysis of the court decisions and precedents. Two forms of rationality of law are opposed: one based on custom and knowledge of which is an “artificial reason”, the other based on natural law and the prescriptions of the judges’ individual reason. Naturalism is perhaps the most pervasive “-ism” in contemporary philosophy. Different variations of naturalism can be found in virtually all corners of theoretical and practical philosophy. Critics have rightfully noted that it is (a) often not clear what “naturalism” means exactly and, subsequently, (b) whether those who consider themselves naturalists in the same philosophical debate actually hold compatible, let alone the same, beliefs. -/- Among the different forms of naturalism that hold currency today, scientific naturalism seems to be the most (...) widespread and therefore articulated. It can be summarized as the ontological thesis that what exists are solely the entities posited by the natural sciences and the methodological thesis that philosophical inquiry should take the results of the natural sciences as authoritative. The paradigm of scientific naturalism has received a growing amount of critical attention from within and outside of analytic philosophy in the last few decades. The main thrust of a host of these critiques is to “soften” the claim of scientific naturalism by rejecting its reductionist or even eliminativist implications, yet retaining a commitment to naturalism under a new description. This has led to a proliferation of new forms of naturalism that seek to broaden the criteria which determine what belongs to the natural world, usually self-titled as a “liberal” or “relaxed” naturalism. Within the analytic tradition, naturalism has been thus critiqued since the 1990s by thinkers like Hornsby, Strawson, Nagel, Stroud, Putnam, McDowell or more recently Baker, De Caro, Macarthur, Beale & Kidd, Cahill & Raleigh. Such critiques are commonly motivated by Neo-Aristotelian and/or Wittgensteinian intuitions. From outside the analytic tradition, phenomenologists in the Husserlian tradition as well as thinkers in the Idealist tradition have been steadfastly critical of scientific naturalism as well, albeit for different reasons. However, these new proposals all have to contend with scientific naturalism and its more restrictive criteria for what counts as natural (although these are debated in their own right). (shrink)

The Reverend Dr John Beale, FRS, DD, and chaplain to Charles II, carried out a vigorous campaign in the early Royal Society for the reform of agriculture, trade, and public education-reforms which signalled his continuing commitment to the ideas not only of Bacon, but of Hartlib and Comenius as well. In addition to promoting orchard plantations and expanded commercial horticulture, he collaborated with Evelyn, Oldenburg, and Houghton to publish or publicize items on the improvement of agriculture and the national (...) economy. His later writings, including the unfinished manifesto ‘From Utopia’, also identify the ‘mercantilist’ thought of Thomas Mun as a crucial link in Beale's alignment of technology, socioeconomic reform, and the religious and ethical values of Christian humanism. This paper investigates the emergence, in the second part of the 17th century, of a new body of experimental knowledge dealing with the chemical transformations of water taking place in plants. We call this body of experimental knowledge a “chemical history of vegetation.” We show that this chemical natural history originated, in terms of recipes and methods of investigation, in the works of Francis Bacon and that it was constructed in accordance with Bacon's precepts for putting together natural and experimental (...) histories. Our paper covers a wide array of experimental investigations, carried out by people with different backgrounds, theoretical assumptions, and metaphysical allegiances (Thomas Browne, Robert Sharrock, Robert Boyle, Nehemiah Grew, John Beale, and John Evelyn). We claim that, despite their differences, these naturalists had a lot in common. First, they treated plants as laboratories of chemical investigation. Second, they used the experiments with plants to develop tools, hypotheses, and operational concepts, which travelled from one naturalist to the other, even when their respective explanatory vocabularies were widely different. Third, we show that some of them also manifested an interest in defining and clarifying the limits and structure of this new body of experimental knowledge, displaying, thus, a certain disciplinary unity. This is a book that contrary to common practice, shows the commonalities of ancient and modern theories of freedom, law, and rational actions. Studying the works of the ancients is necessary to understanding those that follow. Thomas Chaimowicz challenges current trends in research on antiquity in his examination of Montesquieu's and Burk's path of inquiry. He focuses on ideas of balance and freedom. Montesquieu and Burke believe that freedom and balance are closely connected, for without balance within a state (...) there can be no freedom. When Montesquieu speaks of republics, he means those of antiquity as they were understood in the eighteenth century. In this view, freedom can develop only within the framework of established tradition. Edmund Burke's greatest service to political thought may lie in making use of this idea when he fought against the abstractions of the French Revolutionaries. Antiquity as the Source of Modernity examines Montesquieu's "Roman mind," meaning not an attitude influenced by the ancients, but one primarily influenced by Roman heritage. It speaks to the antithesis of monarchy and despotism in Montesquieu's thought and the influence of Tacitus and Pliny the Younger on him. The separation of powers and its relation to the concept of the mixed constitution as well as Montesquieu's smaller masterpiece Considerations on the Causes of the Grandeur and Decadence of the Romans are examined in detail. Finally, the discussion leads seamlessly to Burke, who, as a critical admirer of Montesquieu, partly incorporated his interpretation of the English constitution into his own thinking threatened by teachings of the French Revolution and its British adherents. The central idea of Antiquity as the Source of Modernity is timeless. It is that the ancient past can lead to a clearer understanding of what follows. This perspective represents a reversal of the conventional procedures for conducting this kind of research, but it is a reversal that Chaimowicz embraces in order to add a new dimension to the study and impetration of both Montesquieu and Burke. Thomas Chaimowicz was a distinguished visiting professor at the International Academy of Philosophy in the Principality of Liechtenstein and honorary professor of Roman Lay at the University of Salzburg. Late in a career of more than sixty years, Thomas Burch, an internationally known social demographer, undertook a wide-ranging methodological critique of demography. This open access volume contains a selection of resulting papers, some previously unpublished, some published but not readily accessible [from past meetings of The International Union for the Scientific Study of Population and its research committees, or from other small conferences and seminars]. Rejecting the idea that demography is simply a branch of applied statistics, his work (...) views it as an autonomous and complete scientific discipline. When viewed from the perspective of modern philosophy of science, specifically the semantic or model-based school, demography is a balanced discipline, with a rich body of techniques and data, but also with more and better theories than generally recognized. As demonstrated in this book, some demographic techniques can also be seen as theoretical models, and some substantive/behavioral models, commonly rejected as theory because of inconsistent observations, are now seen as valuable theoretical models, for example demographic transition theory. This book shows how demography can build a strong theoretical edifice on its broad and deep empirical foundation by adoption of the model-based approach to science. But the full-fruits of this approach will require demographers to make greater use of computer modeling [both macro- and micro-simulation], in the statement and manipulation of theoretical ideas, as well as for numerical computation. The Life of Wisdom in Rousseau's "Reveries of the Solitary Walker" is the first complete exegesis and interpretation of Rousseau's final and culminating work, showing its full philosophic and moral teaching. The Reveries has been celebrated as a work of literature that is an acknowledged acme of French prose writing. Thomas L. Pangle argues that this aesthetic appreciation necessitates an in-depth interpretation of the writing's complex and multileveled intended teaching about the normatively best way of life—and how essential this (...) is for a work that was initially bewildering. Rousseau stands out among modern political philosophers in that he restored, to political philosophy, what Socrates and his students (from Plato and Xenophon through Aristotle and the Stoics and Cicero) had made central—and that the previous modern, Enlightenment philosophers had eclipsed: the study of the life and soul of the exemplary, independent sage, as possessor of "human wisdom." Rousseau made this again the supreme theme and source of norms for political philosophy and for humanity's moral as well as civic existence. In his analysis of The Reveries, Pangle uncovers Rousseau's most profound exploration and articulation of his own life, personality, soul, and thought as "the man of nature enlightened by reason." He describes, in Rousseau's final work, the fullest embodiment of the experiential wisdom from which flows and to which points Rousseau's political and moral philosophy, his theology, and his musical and literary art. PurposeAlthough there is much public talk about privacy, it seems that there is no definite answer; rather, ambiguous concepts of what privacy is and what indeed privacy in peril is. The overall aim of this paper is to clarify how privacy is defined in the academic literature, what the different concepts of privacy have in common, what distinguish them from one another, and what advantages and disadvantages such definitions have in order to clarify if there is a gap in the (...) existing literature.Design/methodology/approachThis contribution constructs theoretically founded typologies in order to systemize the existing literature of privacy studies and to analyse examples of privacy (threats). Therefore, it mainly is a theoretical approach combined with illustrative examples.FindingsThis paper contains a systematic discussion of the state of the art of privacy studies by establishing a typology of existing privacy definitions and discussing commonalties and differences. In this paper, it is argued that the existing literature is insufficient for studying privacy. Therefore, a critical contribution to privacy studies is needed.Originality/valueA critical contribution to privacy studies avoids pitfalls of the existing literature and strives for the development of theoretical and empirical research methods in order to focus on privacy in the context of domination, asymmetrical power relations, resource control, social struggles, and exploitation. Mais le problème est de distinguer entre Gassendi et ses disciples contemporains de Locke comme source immédiate d'inspiration pour celui-ci. Ses Commonplace Books attestent que Locke avait lu Gassendi avec attention, et son Journal indique que pendant ses séjours à Paris, il fut en contact avec des gassendistes tels Bernier et Launay, dont il acheta les oeuvres pour les emporter (...) en Angleterre. L'analyse que Locke fournit de l'espace est d'un intérêt particulier, car tout en témoignant de l'orientation que Leibniz attribuait à Locke, elle s'écarte nettement des vues de Gassendi sur la question. Although interest in his life and work has grown enomrously in recent years, this is the first complete edition of his correspondence. The texts of the letters are richly supplemented with explanatory notes and full biographical and bibliographical information. This landmark publication sheds new light in abundance on the intellectual life of a major thinker.

Not only our conscious expectations, wishes and intentions are directed towards the future, but also pre- or unconscious tendencies, hunches and anticipations. Using a term coined by Ernst Bloch, they can be summarized as the not-yet-conscious. This not-yet-consciousness usually unfolds spontaneously and without a plan; it is not expected or striven for, but rather emerges in consciousness in such a way that the subject is surprised by itself, so to speak. This gives rise to phenomena such as the conspicuous, the (...) accidental, the new and the improvised, which are particularly important for understanding creative and therapeutic processes. The aim of this article is to develop a general phenomenological framework for understanding not-yet-conscious dynamics and their unfolding in different contexts. To this end, the structure of the not-yet-conscious is examined in the light of protention, in which the phenomenon is fundamentally located. (shrink)
