Cellular 3D structures, for example, organoids, are an excellent model for studying and developing treatments for various diseases, including hereditary ones. Therefore, they are increasingly being used in biomedical research. From the point of view of safety and efficacy, recombinant adeno‐associated viral (rAAV) vectors are currently most in demand for the delivery of various transgenes for gene replacement therapy or other applications. The delivery of transgenes using rAAV vectors to various types of organoids is an urgent task, however, it is (...) associated with a number of problems that are discussed in this review. Cellular heterogeneity and specifics of cultivation of 3D structures determine the complexity of rAAV delivery and are sometimes associated with low transduction efficiency. This review surveys the main ways to solve emerging problems and increase the efficiency of transgene delivery using rAAVs to organoids. A clear understanding of the stage of development of the organoid, its cellular composition and the presence of surface receptors will allow obtaining high levels of organoid transduction with existing rAAV vectors. Recombinant adeno‐associated viruses (rAAVs) are promising vectors for the delivery of various genetic constructs into eukaryotic cells. rAAVs have a number of properties that make it possible to successfully use them both in vitro and in vivo. Purification and concentration of rAAV vectors are critical for achieving high viral titer, stability, efficiency, and purity. This review systematically analyses all available purification approaches. The purification methods described in this work differ substantially from each other in mechanisms, efficiency, labor time, and cost. (...) Historians of Russian literature have often succumbed to this temptation and it became almost commonplace to read that in Russia the Realism of the nineteenth century gave way to the Symbolism which began in the twentieth; that Symbolism gave way to Futurism; and that in due course all the earlier “-isms” were swept aside by the Socialist Realism of the Soviet period. For Marxist-oriented Soviet [End Page 371] historians such preoccupation with compartmentalization was obsessive. This makes more than welcome a series of essays which is at pains to emphasize the essential connections and kinship between these various “-isms.”Four of the seven essays are intended to demonstrate how the Symbolists, while ostensibly breaking with the earlier tradition of positivism (the idea that art’s essential purpose is to help build a better world), were, in a number of important respects, continuing and even reinforcing what they claimed to despise. Thus, regarding art and life as indissolubly linked, they attempted to turn life into art and art into life. Michael Wachtel shows how Viacheslav Ivanov attempted to overcome his grief at the death of his first wife by using his creative powers to make contact with her “beyond the grave.”Irene Masing-Delic traces the Pygmalion myth through Pushkin, Baratynsky, Turgenev, and Chernyshevsky to the Symbolists who were especially attracted by a myth which showed how an artist could create life and even conquer death. In “Andrei Bely and the Argonauts’ mythmaking,” Alexander Lavrov considers the role of the “Argonaut” circle in establishing a Russian Symbolist culture in the first years of the twentieth century. The Argonauts’ credo was “life-creation” rather than “art-creation,” and they claimed to make no distinction between “talent for living” and “talent for writing.” Lavrov argues that the failure of the relationship between Bely and Nina Petrovskaia marked the disintegration of “Argonautism” and the culture they sought to create. It is left to Irina Gutkin in the concluding essay to guide us through the grotesqueries of Futurism into the apparently diametrically opposed, sanitized world of Socialist Realism, by way of Constructivism, Marxism, and Meyerholdian bio-mechanics, leaving us in little doubt that, appearances to the contrary, each of these movements was concerned with turning art into life and using it to create a better world. So ultimately, as she neatly sums up the thesis of the whole collection, “The aesthetic utopian ideal of art creating life, which took shape at the beginning of the twentieth century, was fused with the utilitarian tradition of the 1860s” (p. 181).I have deliberately reserved for the end comment on Irina Paperno’s introduction and opening article (“The Meaning of Art: Symbolist Theories”) in order to emphasize the manifest contribution she and her co-editor, Joan Delaney Grossman, have made to ensuring the coherence of this admirable collection. Professor Paperno’s clear and concise coverage of her topics makes it evident that she was determined that the book should not turn into the kind of bits-and-pieces patchwork that too many publications of this sort tend to [End Page 372] become without a firm editorial hand at the helm. It is well known that the dominant paradigm underlying the free movement jurisprudence of the Court of Justice has progressively shifted from the ‘market citizen’ to the EU citizen: in order to invoke free movement and equal treatment rights an economic nexus is no longer needed, allowing, for example, students to claim equal treatment with host Member State nationals as regards access to education and the receipt of study grants. Normatively, however, this raises important questions. The classic logic whereby these (...) free movement and equal treatment rights were provided to Member State nationals as a quid pro quo for their contribution to and participation in a project of European integration—the establishment of the common market—is undermined. A new ‘aim’ or ‘rationale’ is thus needed, capable of explaining and informing the citizenship jurisprudence of the Court. The purpose of this article is to construct a conceptual framework for the Court’s EU citizenship jurisprudence by relying on a cosmopolitan vision of citizenship as set out by Immanuel Kant in his seminal work Zum ewigen Frieden. It is submitted in this article that Kant’s cosmopolitan right of hospitality, meant to guarantee interaction between the peoples of the world with a view to developing a shared universal conscience characterised by a desire for peace and prosperity, in essence informs the EU citizenship jurisprudence of the Court. Violent video games (VVGs) are a source of serious and continuing controversy. They are not unique in this respect, though. Other entertainment products have been criticized on moral grounds, from pornography to heavy metal, horror films, and Harry Potter books. Some of these controversies have fizzled out over time and have come to be viewed as cases of moral panic. Others, including moral objections to VVGs, have persisted. The aim of this paper is to determine which, if any, of the (...) concerns raised about VVGs are legitimate. We argue that common moral objections to VVGs are unsuccessful, but that a plausible critique can be developed that captures the insights of these objections while avoiding their pitfalls. Our view suggests that the moral badness of a game depends on how well its internal logic expresses or encourages the players’ objectionable attitudes. This allows us to recognize that some games are morally worse than others—and that it can be morally wrong to design and play some VVGs—but that the moral badness of these games is not necessarily dependent on how violent they are. Physicians have special professional obligations to respond to medical emergencies. A bioterrorism attack would be a medical emergency. Thus, it seems that physicians would have an obligation to respond to a bioterrorist attack. However, the scope of those obligations, and their limits, are vexed topics. General rules may be comforting but the details and nuances of particular situations will always be relevant.

This paper concerns a feature of the deontic landscape that, although pervasive, has hitherto escaped notice. None of the current categorizations of duties adequately capture a common and important form of duty, the “open duty.” The difference between open and closed duties, whether perfect or imperfect, rests not on the side of the end or action enjoined by the duty, but on that of the agents who are enjoined to act. An open duty belongs to more than one person, not (...) all of whose performance of the action is required to fulfill the duty. An example would be the situation faced by American doctors following Hurricane Katrina, when it was imperative that some of them provide aid, but when it was just as necessary that many remain where they were to treat local patients. This article explores the emerging direction of business strategy as expressed by the concept of evolutionary advantage. It examines first the major forms of business knowledge over the last century, then considers mainstream frameworks for strategic analysis, and offers, as a compelling alternative, the emerging notions of evolutionary development and evolutionary learning. This article analyses how Rousseau's First Discourse and the questions it posed about human progress and the reform of society were debated in the institutional context of the Berlin Academy by Formey and Herder. Despite some important disagreements, Formey and Herder fundamentally shared Rousseau's assumption that erudition could be detrimental both to society and to the individual. In order to limit the socially corrosive effects of the arts and the sciences, and in an attempt to realize their full beneficent potential, (...) they called for their reform through institutional regulation and management by a meritocracy of scholarly experts. Drawing on Hume, Herder in particular developed a positive role for the modern state as an active agent of enlightenment, provided it not only promoted the arts and sciences but also guaranteed freedom and the rule of law to ensure their flourishing. First-order formalisations are often preferred to propositional ones because they are thought to underwrite the validity of more arguments. We compare and contrast the ability of some well-known logics—these two in particular—to formally capture valid and invalid arguments. We show that there is a precise and important sense in which first-order logic does not improve on propositional logic in this respect. We also prove some generalisations and related results of philosophical interest. The rest of the article investigates the results’ philosophical (...) significance. A first moral is that the correct way to state the oft-cited superiority of first-order logic vis-`a-vis propositional logic is more nuanced than often thought. The second moral concerns semantic theory; the third logic’s use as a tool for discovery. A fourth and final moral is that second-order logic’s transcendence of first-order logic is greater than first-order logic’s transcendence of propositional logic. (shrink)

The term ‘Orthodox Christian medicine’ has become so customary among believers in Russia that the concept behind it is rarely questioned. However, it is worth articulating both for the sake of the profession and for the benefit of patients whether such a phrase is at all meaningful and if so what exactly it stands for. In simplified form, the argument that I am defending holds that the incompatibility of our freedom with determinism follows from the conjunction of (1) a plausible supervenience claim which says that whether a human agent is free depends only on what happens during the agent’s life and (2) a freedom-cancellation principle of Richard Gale which says that an agent is not free if all of her actions are intentionally brought about by another agent. The article shows that Kuhn's concept of incommensurability emphasizes mainly the objective dimension of communication. To the thesis about the incommensurability of the meanings of scientific concepts in competing paradigms, we oppose the idea of a three-dimensional space of communicative dimensions. We supplement the objective dimension of communication, within which the environmental evolutionary selection of the best knowledge is carried out, with equal social and temporal horizons.
