Insulators play a central role in subdividing the chromosome into a series of discrete topologically independent domains and in ensuring that enhancers and silencers contact their appropriate target genes. In this review we first discuss the general characteristics of insulator elements and their associated protein factors. A growing collection of insulator proteins have been identified including a family of proteins whose expression is developmentally regulated. We next consider several unexpected discoveries that require us to completely rethink how insulators function (and (...) how they can best be assayed). These discoveries also require a reevaluation of how insulators might restrict or orchestrate (by preventing or promoting) interactions between regulatory elements and their target genes. We conclude by connecting these new insights into the mechanisms of insulator action to dynamic changes in the three‐dimensional topology of the chromatin fiber and the generation of specific patterns of gene activity during development and differentiation. In 2008, researchers created human three-dimensional neural tissue – known as the pioneering work of “brain organoids.” In recent years, some researchers have transplanted human brain organoids into animal brains for applicational purposes. With these experiments have come many ethical concerns. It is thus an urgent task to clarify what is ethically permissible and impermissible in brain organoid research. This paper seeks (1) to sort out the ethical issues related to brain organoid research and application and (2) to propose future (...) directions for additional ethical consideration and policy debates in the field. Toward (1), this paper first outlines the current state of brain organoid research, and then briefly responds to previously raised related ethical concerns. Looking next at anticipated scientific developments in brain organoid research, we will discuss (i) ethical issues related to in vitro brain organoids, (ii) ethical issues raised when brain organoids form complexes or have relationships with other entities, and (iii) ethical issues of research ethics and governance. Finally, in pursuit of (2), we propose research policies that are mindful of the ethics of brain organoid research and application and also suggest the need for an international framework for research and application of brain organoids. In this paper, we outline how one might conduct a comprehensive ethical evaluation of human brain organoid transplantation in animals. Thus far, ethical concerns regarding this type of research have been assumed to be similar to those associated with other transplants of human cells in animals, and have therefore not received significant attention. The focus has been only on the welfare, moral status, or mental capacities of the host animal. However, the transplantation of human brain organoids introduces several new ethical (...) issues. Many of these are related to uncertainty regarding whether or not brain organoids might be conscious. While these concerns might not be immediately relevant, they warrant closer scrutiny. We discuss how various ethical issues are relevant to different stages of human brain organoid transplantation and can guide the ethical evaluation of research. Our examination would broaden the horizons of the debate on the transplantation of brain organoids. (shrink)

The formation and recall of memories are fundamental aspects of life and help preserve the complex collection of experiences that provide us with a sense of identity and autonomy. Scientists have recently started to investigate pharmacological agents that inhibit or “dampen” the strength of memory formation and recall. The development of these memory-dampening agents has been investigated for the treatment of posttraumatic stress disorder (PTSD). Currently, these agents are being tested in multicenter clinical trials and will likely soon be approved (...) for the treatment of PTSD. With advancements in technology, more targeted memory-dampening techniques may be developed in the future. Accessibility to these agents will inevitably affect one's sense of identity and also one's sense of autonomy. Therefore, it is essential that the legal and ethical implications of using these agents be examined for governments and courts to appropriately address issues that may emerge. (shrink)

The infamous difficulty of Lacan's writing has its own, very apt synecdoche: the matheme. What makes this ‘little letter’ that structures the signifier so apposite a device is how it stymies even those sophisticated readers for whom Lacan is as close-readable as Mallarm e. The proposition offered here is that this crisis of reading is not the consequence of either some terrible mistake or egregious failing in character, but rather a typically Lacanian move by which his text stages the very (...) theory it is presumably explicating. This article suggests that by striving to read the matheme, instead of seizing upon excuses to avoid it, we can begin to understand what Lacan meant when he said that everything emerges from the structure of the signifier. Quelle est la nature de la liberté réelle (réelle) dans notre société? Je présenterai ici une nouvelle théorie du libéralisme que j’appelle « libéralisme orienté vers la croissance ». D’abord, j’examine le concept de liberté positive et négative d’Isaiah Berlin et je pose la question fondamentale en matière de liberté, à savoir que la liberté est un idéal paradoxal. J’identifierai deux paradoxes : l’un porte sur la liberté ordinaire et les valeurs sophistiquées, l’autre sur la question de la libération et (...) de l’apathie des individus ou des populations. Comment pouvons-nous surmonter ces paradoxes? Pour répondre à cette question, j’examinerai l’argument de Charles Taylor sur la liberté positive (liberté) et en élargirai les implications. Le paradoxe est d’autant plus lorsqu’il s’agit de communautés, comme la société japonaise. Au-delà du mode de pensée communautaire de Taylor, par exemple, je construis une nouvelle théorie de la liberté en proposant de suivre trois principes : le principe de la vertu (estime de soi), le principe du changement générateur et le principe de la différenciation. In making his comparative analysis of the Japanese system, Professor Aoki critically examines conventional notions about the microstructure of the market economy that have strongly shaped and influenced economists' approach to industrial organization. While these (...) notions may constitute an appropriate foundation for the analysis of the highly market-oriented Western economies, the author has found that a more complete understanding of the Japanese economy requires us to broaden such 'specific' notions. Recent fMRI studies have shown that functional networks can be extracted even from resting state data, the so called “resting state networks” (RSNs) by applying independent component analysis (ICA). However, compared to fMRI, EEG and MEG have much higher temporal resolution and provide a direct estimation of cortical activity. To date, MEG studies have applied ICA for separate frequency bands only, disregarding cross-frequency couplings. In this study, we aimed to detect EEG-RSNs and their interactions in all frequency bands. We applied (...) low resolution brain electromagnetic tomography-ICA (LORETA-ICA) to resting-state EEG data in 80 healthy subjects using five frequency bands (delta, theta, alpha, beta and gamma band) and found five RSNs in alpha, beta and gamma frequency bands. Next, taking into account these frequency properties, five RSNs were identified; 1) the visual network, 2) dual-process of visual perception network, characterized by a negative correlation between the right ventral visual pathway (VVP) and left posterior dorsal visual pathway (DVP), 3) self-referential processing network, characterized by a positive correlation between the medial PFC and right VVP, 4) dual-process of memory perception network, functionally related to a negative correlation between the left VVP and the precuneus and 5) sensorimotor network. To detect aging-related changes of these five RSNs, the subjects were divided into three age groups: younger, middle aged, and elderly group, and Student's t test with Bonferroni correction on each coefficient of five independent components were performed. We found a significant attenuation in dual-process of visual perception network in elderly relative to middle aged subjects. Overall findings indicate that LORETA-ICA with EEG data can precisely identify five RSNs in their intrinsic frequency bands, and correct correlations and aging-related changes between and within RSNs. This paper highlights the need to apply an equity lens when assessing the impact of preemption and related legal doctrines on community health. Community autonomy to set and pursue public health priorities is an essential part of achieving health equity. Unfortunately, the priorities of organized industry interest groups often conflict with health equity goals. These groups have a history of successfully using law to limit community autonomy to pursue public health measures, most notably through preemption and related legal doctrines. We (...) examine this phenomenon using two examples. First, we look at dental association efforts to block the spread of dental therapists in Indian Country. Native dental therapists have been improving the oral health of native people in Alaska for over a decade; yet the national and state dental associations have sought with mixed success to leverage state and federal laws to hinder other tribal governments from utilizing these providers. We compare these efforts with a restaurant association-led movement to limit what municipal governments can do to address food-based health inequities, focusing on a “model” ALEC bill that started in Ohio. Finally, we discuss how municipalities and Tribes are fighting back and continuing to pursue health equity for their communities. (shrink)

This study examined the effects of mood and risk-taking on divergent and convergent thinking using a Q-learning computation model. The results revealed that while mood was not significantly related to divergent or convergent thinking (as creative thinking types), risk-taking exerted positive effects on divergent thinking in the face of negative rewards. The results were consistent with the representational change theory in insight problem solving. Although this theory accounts directly for insight, the underlying idea of going beyond current contexts and implicit (...) constrains could be applied to creative thinking as well. The results indeed accounted for the relevance of this theory to divergent thinking. The current study is one of the first empirical studies simultaneously examining the role of mood and risk-taking in creativity. In particular, no related studies exist that took a computational approach to estimate the relevant parameters in the framework of dynamic optimization. Our Q learning model enables to distinguish and identify the different roles of mood and risk-taking in updatingQvalues and making decisions. (shrink)

The role of government in East Asian economic development has been a contentious issue. Two competing views have shaped enquiries into the source of the rapid growth of the high-performing Asian economies and attempts to derive a general lesson for other developing economies: the market-friendly view, according to which government intervenes little in the market, and the developmental state view, in which it governs the market. What these views share in common is a conception of market and government as alternative (...) mechanisms for resource allocation. They are distinct only in their judgement of the extent to which market failures have been, and ought to be, remedied by direct government intervention. This collection of essays suggests a breakthrough, third view: the market-enhancing view. Instead of viewing government and the market as mutually exclusive substitutes, it examines the capacity of government policy to facilitate or complement private sector co-ordination. The book starts from the premiss that private sector institutions have important comparative advantages over government, in particular in their ability to process information available on site. At the same time, it recognizes that the capabilities of the private sector are more limited in developing economies. The market-enhancing view thus stresses the mechanisms whereby government policy is directed at improving the ability of the private sector to solve co-ordination problems and overcome other market imperfections. In presenting the market-enhancing view, the book recognizes the wide diversity of the roles of government across various East Asian economiesincluding Japan, Korea, Hong Kong, Malaysia, and China and its path-dependent and developmental stage nature. The intermediate logics have been classified into slices (cf. Hosoi [1]), but the detailed structure of slices has been studied only for the first two slices (cf. Hosoi and Ono [2]). In order to study the structure of slices, we give a method of a finer classification of slices & n (n 3). Here we treat only the third slice as an example, but the method can be extended to other slices in an obvious way. It is proved that each (...) subslice contains continuum of logics.
