Synthetic biology is a research field that has grown rapidly and attracted considerable attention. Most prominently, it has been labelled the ‘engineering of biology’. While other attempts to label the field have been also pursued, the program of engineering can be considered the core of the field’s disciplinary program, of its identity. This article addresses the success of the ‘engineering program’ in synthetic biology and argues that its success can partly be explained by distinct practices of persuasion that aim at (...) persuading scientific, but also non-scientific audiences. The article explores two different modes of persuasion:, building tools as heuristic models and posing visionary claims. Objects such as the toggle switch or the synthetic oscillator in synthetic biology can more adequately be described as heuristic models of engineering instead of simply as prototypes of ‘tools’. Posing visionary claims can be also understood as a persuasion practice, since the claims are used to construct the societal relevance of the field. Drawing upon Michel Callon’s ‘sociology of translation’, I argue that both practices of persuasion aim at ‘enrolling’ entities into the disciplinary identity. The article is based on the textual analysis of rhetorical practices in three synthetic biology review articles which are considered seminal for the history of the field. (shrink)

The precise orchestration of two opposing protein complexes – one in the cytoplasm (β‐catenin destruction complex) and the other at the plasma membrane (LRP6 signaling complex) – is critical for controlling levels of the transcriptional co‐factor β‐catenin, and subsequent activation of the Wnt/β‐catenin signal transduction pathway. The Wnt pathway component Axin acts as an essential scaffold for the assembly of both complexes. How the β‐catenin destruction and LRP6 signaling complexes are modulated following Wnt stimulation remains controversial. A recent study in (...) Science by He and coworkers reveals an underlying logic for Wnt pathway control in which Axin phosphorylation toggles a switch between the active and inactive states. This mini‐review focuses on this and two other recent studies that provide insight into the initial signaling events triggered by Wnt exposure. We emphasize regulation of the β‐catenin destruction and LRP6 signaling complexes and propose a framework for future work in this area. (shrink)

Mammalian synthetic biology holds the promise of providing novel therapeutic strategies, and the first success stories are beginning to be reported. Here we focus on the latest generation of mammalian transgene control devices, highlight state‐of‐the‐art synthetic gene network design, and cover prototype therapeutic circuits. These will have an impact on future gene‐ and cell‐based therapies and help bring drug discovery into a new era. The inventory of biological parts that are essential for life on this planet is becoming increasingly complete. (...) The postgenomic era has provided encyclopedic information on gene‐function correlations and systems biology is now delivering comprehensive details on the dynamics of biochemical reaction networks in living organisms. The time has come to reassemble these catalogued items and design new biological devices and systems with novel and useful functionality in a rational and systematic manner. This is the premise of synthetic biology, a constructive systems biology discipline likely to become the life science revolution of the 21st century. (shrink)

X‐chromosome inactivation ensures dosage compensation between the sexes in mammals by randomly choosing one out of the two X chromosomes in females for inactivation. This process imposes a plethora of questions: How do cells count their X chromosome number and ensure that exactly one stays active? How do they randomly choose one of two identical X chromosomes for inactivation? And how do they stably maintain this state of monoallelic expression? Here, different regulatory concepts and their plausibility are evaluated in the (...) context of theoretical studies that have investigated threshold behavior, ultrasensitivity, and bistability through mathematical modeling. It is discussed how a twofold difference between a single and a double dose of X‐linked genes might be converted to an all‐or‐nothing response and how mutually exclusive expression can be initiated and maintained. Finally, candidate factors that might mediate the proposed regulatory principles are reviewed. (shrink)

The “Slicing Problem” is a thought experiment that raises questions for substrate-neutral computational theories of consciousness, including those that specify a certain causal structure for the computation like Integrated Information Theory. The thought experiment uses water-based logic gates to construct a computer in a way that permits cleanly slicing each gate and connection in half, creating two identical computers each instantiating the same computation. The slicing can be reversed and repeated via an on/off switch, without changing the amount of (...) matter in the system. The question is what do different computational theories of consciousness believe is happening to the number and nature of individual conscious units as this switch is toggled. Under a token interpretation, there are now two discrete conscious entities; under a type interpretation, there may remain only one. Both interpretations lead to different implications depending on the adopted theoretical stance. Any route taken either allows mechanisms for “consciousness-multiplying exploits” or requires ambiguous boundaries between conscious entities, raising philosophical and ethical questions for theorists to consider. We discuss resolutions under different theories of consciousness for those unwilling to accept consciousness-multiplying exploits. In particular, we specify three features that may help promising physicalist theories to navigate such thought experiments. (shrink)

A fundamental property of emotional responses is a change in action tendencies that allow the individual to cope with the situation. Most basically, there are two types of behaviour one can switch to when responding emotionally: approach or withdrawal. The present study examined whether the ability to switch to approach or withdrawal depends on the type of behaviour shown before. Using familiar (Experiment 1) and unfamiliar (Experiment 2) neutral stimuli, we first show that switching from approach to withdrawal (...) is generally easier than vice versa. In Experiment 3, we demonstrate that this holds true even when participants respond to emotional stimuli that typically elicit strong approach or withdrawal tendencies. These results indicate that there is a fundamental asymmetry in the ability to switch from approach to withdrawal or vice versa. As shown in Experiment 3, this asymmetry may represent a serious confound in many previous studies examining the link between stimulus valence and associated action tendencies, suggesting that the link between positive stimuli and approach tendencies may be stronger, and the link between negative stimuli and withdrawal tendencies weaker, than previously believed. The contentious question of bilingual processing cost may be recast as a fresh question of code-switching (CS) strategies—quantitative preferences and structural adjustments for switching at particular junctures of two languages. CS strategies are established by considering prosodic and syntactic variables, capitalizing here on bidirectional multi-word CS, spontaneously produced by members of a bilingual community in northern New Mexico who regularly use both languages (Torres Cacoullos and Travis, 2018). CS strategies become apparent by extending the equivalence constraint, which states that bilinguals (...) avoid CS at points of word placement conflict (Poplack, 1980), to examine points of inconsistent equivalence between the languages. Such sites of variable equivalence are junctures where the word strings of the two languages are equivalent only sometimes due to language-internal variable structures. A case in point for the English-Spanish language pair is the boundary between main and complement clauses, where a conjunction occurs always in Spanish but variably in English. The prosodic distancing strategy is to separate the juncture of the two languages. Here the complement clause appears in a different prosodic unit from the main clause, disproportionately as compared both with monolingual benchmarks and with bilinguals' own unilingual English and Spanish. Prosodic distancing serves to mitigate variable equivalence. The syntactic selection strategy is to opt for the variant that is more quantitatively available and more discourse neutral. Here the preference is for the Spanish complementizer que—regardless of main or complement clause language— which is the more frequent option in bilinguals' combined experience in both their languages, whereas English complementizer that is subject to a number of conditioning factors. Syntactic selection serves to restore equivalence at sites of variable equivalence. Discovery of community CS strategies may spur reconsideration of processing cost as a matter of relative difficulty, which will depend on bilinguals' prosodic and syntactic choices at particular CS sites. Formal methods developed for modeling levels of selection problems have recently been applied to the investigation of major evolutionary transitions. We discuss two new tools of this kind. First, the ‘near-variant test’ can be used to compare the causal adequacy of predictively equivalent representations. Second, ‘state-variable gestalt-switching’ can be used to gain a useful dual perspective on evolutionary processes that involve both higher and lower level populations.

Deciphering how DCAFs (DDB1‐CUL4 Associated Factors) modulate a broad spectrum of cellular processes, including cell cycle progression and maintenance of genomic integrity is critical to better understand cellular homeostasis and diseases. Cells contain more than 100 DCAFs that associate with the Cullin‐Ring Ubiquitin Ligase 4 (CRL4) complex that target specific protein substrates for degradation. DCAFs are thought to act as substrate receptors that dictate the specificity of the ubiquitination machinery (“catalytic DCAFs”). However, recent studies have suggested that some DCAFs might (...) play a different role by targeting CRL4 complexes to distinct cellular compartments (“structural DCAFs”). Once localized to their correct cellular domains, these CRLs dissociate from the structural DCAFs prior to their association with other, substrate‐specific catalytic DCAFs. Thus, we propose that DCAF switches can provide a mechanistic basis for the degradation of proteins that regulate cell growth and proliferation at precise points in space and time. (shrink)

Based on content externalism, the question of whether self-knowledge is authoritative or not has launched a real controversy in the philosophy of mind. Boghossian proposed slow switching argument in defense of incompatibility of the two views. This argument has been criticized by some philosophers through different approaches. Vahid is one of them. He claimed that Boghossian's argument appeals to some controversial assumptions without which it cannot achieve its conclusion. Organisations that design and deploy artificial intelligence (AI) systems increasingly commit themselves to high-level, ethical principles. However, there still exists a gap between principles and practices in AI ethics. One major obstacle organisations face when attempting to operationalise AI Ethics is the lack of a well-defined material scope. Put differently, the question to which systems and processes AI ethics principles ought to apply remains unanswered. Of course, there exists no universally accepted definition of AI, and different systems pose different ethical (...) challenges. Nevertheless, pragmatic problem-solving demands that things should be sorted so that their grouping will promote successful actions for some specific end. In this article, we review and compare previous attempts to classify AI systems for the purpose of implementing AI governance in practice. We find that attempts to classify AI systems proposed in previous literature use one of three mental models: _the Switch_, i.e., a binary approach according to which systems either are or are not considered AI systems depending on their characteristics; _the Ladder_, i.e., a risk-based approach that classifies systems according to the ethical risks they pose; and _the Matrix_, i.e., a multi-dimensional classification of systems that take various aspects into account, such as context, input data, and decision-model. Each of these models for classifying AI systems comes with its own set of strengths and weaknesses. By conceptualising different ways of classifying AI systems into simple mental models, we hope to provide organisations that design, deploy, or regulate AI systems with the vocabulary needed to demarcate the material scope of their AI governance frameworks. John Searle and Susanna Siegel have argued that cases of aspect‐switching show that visual experience represents a richer range of properties than colours, shapes, positions and sizes. I respond that cases of aspect‐switching can be explained without holding that visual experience represents rich properties. I also argue that even if Searle and Siegel are right, and aspect‐switching does require visual experience to represent rich properties, there is reason to think those properties do not include natural‐kind properties, such as being a (...) tomato. Theories of embodied cognition hold that the conceptual system uses perceptual simulations for the purposes of representation. A strong prediction is that perceptual phenomena should emerge in conceptual processing, and, in support, previous research has shown that switching modalities from one trial to the next incurs a processing cost during conceptual tasks. However, to date, such research has been limited by its reliance on the retrieval of familiar concepts. We therefore examined concept creation by asking participants to interpret modality-specific compound (...) phrases (i.e., conceptual combinations). Results show that modality switching costs emerge during the creation of new conceptual entities: People are slower to simulate a novel concept (e.g., auditory jingling onion) when their attention has already been engaged by a different modality in simulating a familiar concept (e.g., visual shiny penny). Furthermore, these costs cannot be accounted for by linguistic factors alone. Rather, our findings support the embodied view that concept creation, as well as retrieval, requires situated perceptual simulation. (shrink)

Exploration of immune systems in prokaryotes, such as restriction‐modification or CRISPR‐Cas, shows that both innate and adaptive systems possess programmed cell death (PCD) potential. The key outstanding question is how the immune systems sense and “predict” infection outcomes to “decide” whether to fight the pathogen or induce PCD. There is a striking parallel between this life‐or‐death decision faced by the cell and the decision by temperate viruses to protect or kill their hosts, epitomized by the lysis‐lysogeny switch of bacteriophage (...) Lambda. Immune systems and temperate phages sense the same molecular inputs, primarily, DNA damage, that determine whether the cell lives or dies. Because temperate (pro)phages are themselves components of prokaryotic genomes, their shared “interests” with the hosts result in coregulation of the lysis‐lysogeny switch and immune systems that jointly provide the cell with the decision machinery to probe and predict infection outcomes, answering the life‐or‐death question. In the present study, we use a training design in two experiments to examine whether bilingual language switching facilitates two components of cognitive control, namely monitoring and inhibitory control. The results of Experiment 1 showed that training in language switching reduced mixing costs and the anti-saccade effect among bilinguals. In Experiment 2, the findings revealed a greater decrease of mixing costs and a smaller decrease of the anti-saccade effect from pre- to post-training for the language switching training group compared to (...) the L2 training group. Overall, the results suggest that extensive exercise in monitoring and inhibitory control among languages in an experimental setting may enhance the corresponding components of cognitive control. This article deals with an argument reported by Razi (d. 1210) that attempted to undermine the immaterialist position about human nature. After some introductory remarks and explanation of the conceptual background, the article analyses the structure of the argument, with special attention to the idea of soul-switching.’ Some comparisons are made between the argument reported by Razi and a number of arguments from modern and contemporary eras of philosophy. One section is devoted to the critique of the argument and its (...) conceptual basis. This article shows that the argument reported by Razi is a methodological antecedent of a family of contemporary epistemological arguments against substance dualism. It is also shown that discussion of the argument could be useful to highlight a weakness in some, but not all, versions of immaterialism about human nature. (shrink)

This study investigates switching costs in landmark-based wayfinding using olfactory and visual landmark information. It has already been demonstrated that there seem to be no switching costs, in terms of correct route decisions, when switching between acoustically and visually presented landmarks. Olfaction, on the other hand, is not extensively focused on in landmark-based wayfinding thus far, especially with respect to modality switching. The goal of this work is to empirically test and compare visual and olfactory landmark information with regard to (...) their suitability for wayfinding including a modality switch. To investigate this, an experiment within a virtual environment was conducted in which participants were walked along a virtual route of 12 intersections. At each intersection, landmark information together with directional information was presented, which was to be memorized and recalled in the following phase, either in the same or in the other modality. The results of the study show that, in contrast to the no-switching costs between auditory and visual landmarks in previous studies, switching costs occur when switching modality from visual to olfactory and vice versa. This is indicated by both longer decision times and fewer correct decisions. This means that a modality switch involving olfactory landmark information is possible but could lead to poorer performance. Therefore, olfaction may still be valuable for landmark-based-wayfinding. We argue that the poorer performance in the switching-condition is possibly due to higher cognitive load and the separate initial processing of odors and images in different cognitive systems. Inducing the rubber hand illusion (RHI) requires that participants look at an imitation hand while it is stroked in synchrony with their occluded biological hand. Previous explanations of the RHI have emphasized multisensory integration, and excluded higher cognitive functions. We investigated the relationship between the RHI and higher cognitive functions by experimentally testing task switch (as measured by switch cost) and mind wandering (as measured by SART score); we also included a questionnaire for attentional control that comprises two (...) subscales, attention-shift and attention-focus. To assess experience of RHI, the Botvinick and Cohen (1998) questionnaire was used and illusion onset time was recorded. Our results indicate that rapidity of onset reliably indicates illusion strength. Regression analysis revealed that participants evincing less switch cost and higher attention-shift scores had faster RHI onset times, and that those with higher attention-shift scores experienced the RHI more vividly. These results suggest that the multi-sensory hypothesis is not sufficient to explain the illusion: higher cognitive functions should be taken into account when explaining variation in the experience of ownership for the rubber hand. This article discusses xenotransplantation and examines the way its scientific promoters have defended their technology against potentially damaging public representations. The authors explore the criteria used to legitimate the selection of the pig as the best species from which to “harvest” transplant tissues in the future. The authors’ analysis shows that scientists and medical practitioners routinely switch between scientific and cultural repertoires. These repertoires enable such actors to exchange expert identities in scientific discourse for public identities in cultural discourse. (...) These discourses map onto similarities and differences between animal donors and human hosts. Realists about corporate agency, according to whom corporate agents may have aims above and beyond those of the individuals who make them up, think that individual agents may switch between participating in individual and corporate agency. My aim is, however, to argue that the inescapability of individual agency spells out a difficulty for this kind of switching – and, therefore, for realism about corporate agency. To do so, I develop Korsgaard’s notion of plight inescapability, which on my take suggests (...) that individual agents are continuously faced with fully exercising their individual agency (absent external limits at the time of its exercise). But then individual agents may not switch to acting as members of corporate agents if the aims of the corporation differ from their own, in the sense that they have different mental state tokens. But as it is possible to participate fully in the action of a corporate entity, this incompatibility between individual and corporate aims suggests that we ought to think of corporate agents as in some sense reducible to individual aims. It has been proposed that processing sentential negation recruits the neural network of inhibitory control. In addition, inhibition mechanisms also play a role in switching languages for bilinguals. Since both processes may share inhibitory resources, the current study explored for the first time whether and how language-switching influences the processing of negation. To this end, two groups of Spanish-English bilinguals participated in an encoding-verification memory task. They read short stories involving the same two protagonists, referring to their activities in four (...) different scenarios in Spanish or English. Following each story, the participants received verification questions requiring “yes” or “no” responses depending on whether a given fact was correctly referred to one of the protagonists. Some of the verification questions were in the story’s original language and others in the alternate language. Results revealed that language-switching facilitated negative responses compared to affirmative responses, exclusively for questions switching from dominant language to non-dominant language. This effect might reflect that the domain-general mechanisms of inhibitory control are recruited at least partially for both language switch and negation process simultaneously, although this phenomenon is modulated by language dominance. The use of more than one language in a literary text is called literary multilingualism. Ricci's trilogy presents this linguistic phenomenon as the author uses more than one linguistic code in the same text: English, Italian, and dialect. This work describes how code-switching becomes an important device of mimesis, representing not just a switching in language, but also a switch in culture, style, and in voice. This study demonstrates how Ricci exploited his linguistic richness on a literary level creating (...) novels that present the Italian world in a realistic way, but at the same time making use of linguistic varieties as literary device. The contributions to this volume are concerned with questions of form, function, and genesis of canonical switch-reference systems.

Recent models of the conceptual system hold that concepts are grounded in simulations of actual experiences with instances of those concepts in sensory-motor systems (e.g., Barsalou, 1999, 2003; Solomon & Barsalou, 2001). Studies supportive of such a viewhave shown that verifying a property of a concept in one modality, and then switching to verify a property of a different concept in a different modality generates temporal processing costs similar to the cost of switching modalities in perception. In addition to non-emotional (...) concepts, the present experiment investigated switching costs in verifying properties of positive and negative (emotional) concepts. Properties of emotional concepts were taken from vision, audition, and the affective system. Parallel to switching costs in neutral concepts, the study showed that for positive and negative concepts, verifying properties from different modalities produced processing costs such that reaction times were longer and error rates were higher. Importantly, this effect was observed when switching from the affective system to sensory modalities, and vice-versa. These results support the embodied cognition view of emotion in humans. (shrink)

Genomic imprinting is an epigenetic mechanism resulting in the preferential expression of the maternal or paternal alleles of a specific subset of genes in the mammalian genome. A key but relatively unexplored question is how imprints are established in the germline. New observations(1) on two classical imprinting disorders, the Prader‐Willi (PWS) and Angleman (AS) syndromes, offer the first genetic insight into this process. Molecular analysis of imprinting mutations that interfere with the appropriate establishment of the maternal and paternal epigenotypes has (...) led to the identification of imprinted transcripts that could be involved in switching imprints in the germlines. Studies on event-related potentials (ERP) in code-switching (CS) have concentrated on single-word insertions, usually nouns. However, CS ranges from inserting single words into the main language of discourse to alternating languages for larger segments of a discourse, and can occur at various syntactic positions and with various word classes. This ERP study examined native speakers of Russian who had learned German as a second language; they were asked to listen to sentences with CS from their second language, German, to their (...) first language, Russian. CS included either a whole prepositional phrase or only the lexical head noun of a prepositional phrase. CS at nouns resulted in a late positive complex (LPC), whereas CS at prepositions resulted in a broad early negativity, which was followed by an anterior negativity with a posterior positivity. Only in the last time window (800–1000 ms) did CS at prepositions result in a broad positivity similar to CS at nouns. The differences between both types of CS indicate that they relate to different psycholinguistic processes. (shrink)

Poly(ADP‐ribosyl)ation is a post‐translational modification occurring in the nucleus. The most abundant and best‐characterized enzyme catalyzing this reaction, poly(ADP‐ribose) polymerase 1 (PARP1), participates in fundamental nuclear events. The enzyme functions as molecular “nick sensor”. It binds with high affinity to DNA single‐strand breaks resulting in the initiation of its catalytic activity. Activated PARP1 promotes base excision repair. In addition, PARP1 modifies several transcription factors and thereby precludes their binding to DNA. We propose that a major function of PARP1 includes the (...) silencing of transcription preventing expression of damaged genes. Concomitant stimulation of DNA repair suggests that PARP1 acts as a switch between transcription and DNA repair. Another PARP‐type enzyme, tankyrase, is involved in the regulation of telomere elongation. Tankyrase modifies a telomere‐associated protein and thereby prevents it masking telomeric repeats providing access of telomerase for telomere elongation. Suppose COVID-19 is the runaway tram in the famous moral thought experiment, known as the “Bystander at the Switch.” Consider the two differentiated responses of governments around the world to this new threat, namely the option of quarantine/lockdown and herd immunity. Can we contrast the hypothetical with the real scenario? What do the institutional decisions and strategies for dealing with the virus, in the beginning of 2020, signify in a normative moral framework? This paper investigates these possibilities in order (...) to highlight the similarities and, more importantly, the differences that exist between utilitarianism and Kantian ethics. Analysis shows that the hypothetical scenario can never be fully compared to the complex multifactorial nature of the real world. But if a comparison is attempted, the most obvious difference between the two governmental strategies is the concept of duty within the Kantian perspective. Ultimately, it is a matter of comparing freedom and life. Attributing a moral “priority ticket” to one or the other can be analysed through interpersonal aggregation. (shrink)

Recent reports of “switched” embryos and gametes place already fragile couples in unfortunate circumstances, raise vexing questions regarding such fundamental concepts as parenthood and reproduction, and cast a shadow on the integrity of the infertility industry.

Shortcuts to goals are rewarded by faster attainment and punished by more frequent failure, so selection of the various kinds – heuristics, cached sequences (habits or macros), gut instincts – depends on reward history just like other kinds of choice. The speeds of shortcuts lie on continua along with speeds of deliberation, and these continua have no obvious separation points.

Culture-as-situated cognition theory provides insight into the system 1 monitoring algorithm. Culture provides people with an organizing framework, facilitating predictions, focusing attention, and providing experiential signals of certainty and uncertainty as system 1 inputs. When culture-based signals convey that something is amiss, system 2 reasoning is triggered and engaged when resources allow; otherwise, system 1 reasoning dominates.

L’objectif de la contribution est double. Elle vise, d’une part, le développement d’une typologie d’exemples de code switching (CS) permettant de délimiter le corpus adéquat pour tester différentes hypothèses relatives à la grammaire du CS (Jake et al. 2005, MacSwam 2005, 2008, Liceras et al. 2005, 2008). D’autre part, (...) la quantification des données permet un approfondissement des connaissances sur le phénomène du CS dans des productions précoces. Elle incite à relativiser l’importance des énoncés mixtes comme indicateurs d’une majeure ou mineure compétence grammaticale des enfants bilingues dans le cadre de productions langagières (L1 et L2) en situation de communication habituelle avec d’autres partenaires enfants et adultes. The attempt to model the structure of consciousness in split-brain subjects is ongoing. This paper concerns the recently proposed ?switch model? of split-brain consciousness, according to which a split-brain subject possesses only a single stream of consciousness, unified at and across time, that shifts from one hemisphere to the other from moment to moment. The paper argues that while the central explanatory element of the switch model may account for some aspects of split-brain consciousness, the best general picture (...) of split-brain consciousness is still offered by some version of the ?conscious duality model.?
